چکیده
ﻛﻠﺰا ﺑﻪ ﻋﻨﻮان ﮔﻴﺎه روﻏﻨﻲ، ﻫﻤﭽﻮن دﻳﮕﺮ ﻣﺤﺼﻮﻻت زراﻋﻲ ﻣﺘﺎﺛﺮ از ﺗﻨﺶﻫﺎی ﻣﺨﺘﻠﻒ از ﺟﻤﻠﻪ ﺷﻮری ﻣﻲﺑﺎﺷﺪ. ﺑﺎﻛﺘﺮی Pseudomonas fluorescens در ﻫﻤﻴﺎری ﺑﺎ ﮔﻴﺎﻫﺎن ﺑﺎﻋﺚ اﻓﺰاﻳﺶ رﺷﺪ و ﻫﻤﭽﻨﻴﻦ اﻓﺰاﻳﺶ ﻣﻘﺎوﻣﺖ ﮔﻴﺎه در ﺑﺮاﺑﺮ ﻋﻮاﻣﻞ ﺑﻴﻤﺎریزا و ﺗﻨﺶﻫﺎی ﻏﻴﺮزﻳﺴﺘﻲ ﻣﻲﮔﺮدد. در اﻳﻦ ﺗﺤﻘﻴﻖ ﺷﻮری در دو ﺳﻄﺢ ﺻﻔﺮ و 300 ﻣﻴﻠﻲ ﻣﻮﻻر ﻧﻤﻚ NaCl اﻋﻤﺎل ﺷﺪ. ﺣﻀﻮر و ﻋﺪم ﺣﻀﻮر ﺑﺎﻛﺘﺮی ﺑﻪ ﻋﻨﻮان ﻓﺎﻛﺘﻮرﻫﺎی دﻳﮕﺮ اﻋﻤﺎل ﮔﺮدﻳﺪ و ﺑﺮای ﺗﺠﺰﻳﻪﻫﺎی آﻣﺎری از آزﻣﺎﻳﺶ ﻓﺎﻛﺘﻮرﻳﻞ دو ﻋﺎﻣﻠﻲ ﺑﺎ ﻃﺮح ﭘﺎﻳﻪی ﻛﺎﻣﻼ ﺗﺼﺎدﻓﻲ اﺳﺘﻔﺎده ﺷﺪ. ﺑﻪ ﻣﻨﻈﻮر ﺑﺮرﺳﻲ ﭘﺮوﺗﺌﻴﻦﻫﺎی دﺧﻴﻞ در ﺑﻬﺒﻮد ﻣﻘﺎوﻣﺖ ﮔﻴﺎه ﻛﻠﺰا ﺑﻪ ﺗﻨﺶ ﺷﻮری در ﻫﻤﺰﻳﺴﺘﻲ ﺑﺎ ﺑﺎﻛﺘﺮی Pseudomonas fluorescens اﻟﮕﻮی اﻟﻜﺘﺮوﻓﻮرز دو ﺑﻌﺪی ﭘﺮوﺗﺌﻮم رﻳﺸﻪ ﻛﻠﺰا رﻗﻢ Hyola 308 ﻣﻮرد ﺑﺮرﺳﻲ ﻗﺮار ﮔﺮﻓﺖ. ﺑﺪﻳﻦ ﻣﻨﻈﻮر ﻋﺼﺎرهی ﭘﺮوﺗﺌﻴﻨﻲ ﺑﺎﻓﺖ رﻳﺸﻪ ﺑﺎ روش TCA/اﺳﺘﻮن اﺳﺘﺨﺮاج ﮔﺮدﻳﺪ. ژلﻫﺎی ﺣﺎﺻﻞ ﭘﺲ از رﻧﮓ آﻣﻴﺰی ﺑﻪ روش ﻧﻴﺘﺮات ﻧﻘﺮه و ﺗﺼﻮﻳﺮ ﺑﺮداری ﺑﻪ ﻛﻤﻚ ﻧﺮماﻓﺰار ﺑﺎ ژلﻫﺎی ﻧﻤﻮﻧﻪﻫﺎی ﺷﺎﻫﺪ ﻣﻘﺎﻳﺴﻪ و ﺗﻐﻴﻴﺮات ﺑﻴﺎن ﻟﻜﻪﻫﺎی ﭘﺮوﺗﺌﻴﻨﻲ ﻣﻮرد ﺗﺠﺰﻳﻪ و ﺗﺤﻠﻴﻞ ﻗﺮار ﮔﺮﻓﺖ. ﭘﺮوﺗﺌﻴﻦﻫﺎی ﺗﻜﺮارﭘﺬﻳﺮ و ﻣﻌﻨﻲدار در ﺷﺮاﻳﻂ ﺗﻨﺶ ﺑﺎ در ﻧﻈﺮ ﮔﺮﻓﺘﻦ ﻧﻘﻄﻪی اﻳﺰواﻟﻜﺘﺮﻳﻚ و وزن ﻣﻮﻟﻜﻮﻟﻲ ﺷﻨﺎﺳﺎﻳﻲ و در ﺳﻪ ﮔﺮوه ﻋﻤﺪه ﺷﺎﻣﻞ ﭘﺮوﺗﺌﻴﻦﻫﺎی دﺧﻴﻞ در ﻣﺘﺎﺑﻮﻟﻴﺴﻢ، ﺳﻢزداﻳﻲ و ﭘﺮوﺗﺌﻴﻦﻫﺎی ﻧﺎﻗﻞ ﺗﻘﺴﻴﻢ ﺑﻨﺪی ﺷﺪﻧﺪ.

Rapeseed (Brassica napus L.) is an economical oilseed which is affected by salinity stress. Yet,
Pseudomonas fluorescens as a PGPR brings more growth and tolerance against abiotic stress and limiting
conditions for the plant. The proteomic response of genotype Hyola 308 to NaCl salinity (0, and 300 mM
NaCl) was studied in hydroponic culture system. Presence or absence of Pseudomonas fluorescens FY32
was the other factor which is studied. For statistical analysis completely randomized factorial design was
used. 2D-PAGE was applied to determine proteins included in salt tolerance. So protein extract of canola
root was extracted by TCA/acetone method. Resulted gels after silver staining, were pictured and compared
by software. Repeatable spots were identified using PI and experimental Molecular weight. These are
classified into three groups including Proteins involved in metabolism, detoxification and transportation
