چکیده
گندم از مهمترین گیاهان زراعی جهان میباشد. بنابراین ارزیابی واکنش ژنوتیپهای گندم در برابر تنشها ضروری است. در این راستا، اثر تنش خشکی بر میزان فعالیت آنزیم پراکسیداز ز (POX) در ﺳﻪ ﮔﺮوه از ﮔﻨﺪمﻫﺎی ﭘﺎﻳﻴﺰه ﺣﺴﺎس، ﺑﻴﻨﺎﺑﻴﻦ و ﻣﺘﺤﻤﻞ ﺑﻪ ﺧﺸﻜﻲ در ﻣﺮﺣﻠﻪ ﭘﻨﺠﻪزﻧﻲ در ﺳﻪ ﺷﺮاﻳﻂ آﺑﻲ ﻋﺎدی (FC=90)، ﺗﻨﺶ ﻣﺘﻮﺳﻂ (FC=60)و ﺗﻨﺶ ﺷﺪﻳﺪ(FC=30)ﻣﻮرد ﺑﺮرﺳﻲ ﻗﺮار ﮔﺮﻓﺖ. آزﻣﺎﻳﺶ ﺑﻪ ﺻﻮرت ﻓﺎﻛﺘﻮرﻳﻞ ﺑﺎ ﻃﺮح ﭘﺎﻳﻪ ﻛﺎﻣﻼً ﺗﺼﺎدﻓﻲ ﮔﻠﺪاﻧﻲ( در ﺳﻪ ﺗﻜﺮار اﺟﺮا ﺷﺪ و در ﻣﺮﺣﻠﻪ ﭘﻨﺠﻪزﻧﻲ ﻓﻌﺎﻟﻴﺖ آﻧﺰﻳﻢ ﻓﻮق ﺑﺎ اﻟﻜﺘﺮوﻓﻮرز در ژل آﻛﺮﻳﻼﻣﻴﺪ اﻓﻘﻲ 7/5 ﺗﺠﺰﻳﻪ ﮔﺮدﻳﺪ. در اﻳﻦ ﺑﺮرﺳﻲﻫﺎ ﭼﻬﺎر اﻳﺰوزﻳﻢ ﺑﺮای POX، ﺷﻨﺎﺳﺎﻳﻲ ﺷﺪ. ﺗﺠﺰﻳﻪ وارﻳﺎﻧﺲ دادهﻫﺎ ﻧﺸﺎن داد ﻛﻪ ﺗﻨﺶ ﺧﺸﻜﻲ اﺛﺮ ﻣﻌﻨﻲداری در ﻓﻌﺎﻟﻴﺖ ﻛﻠﻴﻪ اﻳﺰوزﻳﻢﻫﺎی ﻣﻮرد ﻣﺸﺎﻫﺪه داﺷﺖ. در ﺳﻪ ﮔﺮوه از ژﻧﻮﺗﻴﭗﻫﺎی ﮔﻨﺪم از ﻟﺤﺎظ ﻓﻌﺎﻟﻴﺖ آﻧﺰﻳﻢﻫﺎی POX1 و POX2، اﺧﺘﻼف ﻣﻌﻨﻲدار ﻣﺸﺎﻫﺪه ﺷﺪ. ﻣﻴﺰان ﻓﻌﺎﻟﻴﺖ آﻧﺰﻳﻤﻲ در ﺗﻨﺶ ﺷﺪﻳﺪ ﺧﺸﻜﻲ ﻧﺴﺒﺖ ﺑﻪ ﺣﺎﻟﺖ ﻋﺎدی اﻓﺰاﻳﺶ ﻳﺎﻓﺖ ﺑﻪ ﻃﻮرﻳﻜﻪ درﺻﺪ اﻓﺰاﻳﺶ ﻓﻌﺎﻟﻴﺖ آﻧﺰﻳﻤﻲ POX4 ،POX3 ،POX2 ،POX1 ﺑﻪ ﺗﺮﺗﻴﺐ 39، 86، 75 و 20 ﺑﻮد. ﺑﺎ وﺟﻮد اﻳﻦ، اﺛﺮ ﻣﺘﻘﺎﺑﻞ ﺗﻨﺶ × ﮔﺮوه ﺑﺮای POX2 و POX3 ﻣﻌﻨﻲدار ﺑﺪﺳﺖ آﻣﺪ. ﺑﻪ ﻃﻮرﻳﻜﻪ ﻣﻴﺎﻧﮕﻴﻦ ﻓﻌﺎﻟﻴﺖ اﻳﻦ آﻧﺰﻳﻢﻫﺎ ﺑﺮای ژﻧﻮﺗﻴﭗﻫﺎی ﮔﺮوه ﺣﺴﺎس ﮔﻨﺪم در ﺳﻪ ﺷﺮاﻳﻂ آﺑﻲ )ﻋﺎدی، ﺗﻨﺶ ﻣﺘﻮﺳﻂ و ﺗﻨﺶ ﺷﺪﻳﺪ( اﺧﺘﻼف ﻣﻌﻨﻲداری ﺑﺎ ﻫﻢ ﻧﺪاﺷﺘﻨﺪ، در ﺣﺎﻟﻴﻜﻪ ژﻧﻮﺗﻴﭗﻫﺎی ﻣﺘﺤﻤﻞ و ﺑﻴﻨﺎﺑﻴﻦ در ﺑﺮاﺑﺮ ﺷﺮاﻳﻂ ﺧﺸﻜﻲ ﭘﺎﺳﺦﻫﺎی ﻣﻌﻨﻲدار و ﻣﺘﻔﺎوﺗﻲ داﺷﺘﻨﺪ.

Wheat is one of the most important crops in the world. So evaluate on of the response of wheat genotypes against stresses is important. So far the effect of drought on the activity of POX enzyme in three group of drought sensitive, intermediate and tolerant winter wheat at three normal (FC=90), intermediate stress (FC=60) and severe stress (FC=30) conditions were studied. The experiment was conducted in factorial form based on completely randomized design in Tabriz university experimental farm. This study identified four isozymes of POX. The analysis of variance showed that the drought stress had a significant effect on the activity of all isozymes studied. Between three groups of wheat genotypes the activities of POX1 and POX2 showed significant difference. Enzyme activity was increased in severe drought stress than normal condition. Percentage of increment in activity for POX1, POX2, POX3 and POX4 were 39, 86, 75 and 20 respectively. However, the drought stress × wheat group interaction for POX2 and POX3 were significant. For susceptible wheat genotypes, difference between three environmental conditions was not significant, but genotype belonging to tolerant wheat group showed significant difference at different conditions.
