چکیده
ﭼﻜﻴﺪه: ﺑﺮرﺳﻲ ﺳﺎﺧﺘﺎر ﺳﻪ ﺑﻌﺪی و ﻋﻮاﻣﻞ ﻣﻮﺛﺮ ﺑﺮ آن ﻣﻲﺗﻮاﻧﺪ درک ﺑﻬﺘﺮی از ﻋﻤﻠﻜﺮد ﭘﺮوﺗﺌﻴﻦﻫﺎ را اراﺋﻪ دﻫﺪ. آﻧﺰﻳﻢ ﭘﺮوﻟﻴﻦ ﻛﺮﺑﻮﻛﺴﻴﻼت رداﻛﺘﺎز ﻣﺮﺣﻠﻪ ﻧﻬﺎﻳﻲ ﺳﻨﺘﺰ ﭘﺮوﻟﻴﻦ را ﻛﺎﺗﺎﻟﻴﺰ ﻣﻲﻛﻨﺪ. ﭘﺮوﻟﻴﻦ ﺑﻪ ﻋﻨﻮان ﻳﻚ اﺳﻤﻮﻻﻳﺖ ﻣﻬﻢ در ﺗﻌﺪﻳﻞ ﻓﺸﺎر اﺳﻤﺰی ﺳﻠﻮلﻫﺎی ﺗﺤﺖ ﺗﻨﺶ ﻫﺎی ﻏﻴﺮ زﻳﺴﺘﻲ ﻧﻘﺶ دارد. ﺑﻪ ﻣﻨﻈﻮر ﻣﻄﺎﻟﻌﻪ اﻳﻦ آﻧﺰﻳﻢ، ﺳﺎﺧﺘﺎر ﺳﻪ ﺑﻌﺪی و ﺗﻮاﻟﻲ آﻣﻴﻨﻮاﺳﻴﺪی ﻣﺮﺑﻮط ﺑﻪ اﻳﻦ آﻧﺰﻳﻢ ﺑﺮای ﮔﻴﺎﻫﺎن Zea
mays, Arabidopsis thaliana, Glycin hispida, Pisum sativum از ﭘﺎﻳﮕﺎهﻫﺎی اﻃﻼﻋﺎﺗﻲ ExPASy وNCBI درﻳﺎﻓﺖ ﺷﺪ. ﺑﺎ اﺳﺘﻔﺎده از اﺑﺰارﻫﺎی ﺑﻴﻮاﻧﻔﻮرﻣﺎﺗﻴﻜﻲ ﺑﺮرﺳﻲﻫﺎ ﺑﺮ روی ﺳﺎﺧﺘﺎرﻫﺎی ﺛﺎﻧﻮﻳﻪ، ﻣﺤﻞ ﺳﺎﺧﺘﺎرﻫﺎی ﺛﺎﻧﻮﻳﻪ، ﻧﻮع و ﻣﻘﺪار اﺳﻴﺪﻫﺎی آﻣﻴﻨﻪ، ﻣﺤﻞ ﻓﺴﻔﻮرﻳﻼﺳﻴﻮن و ﻣﺤﻞ اﺗﺼﺎل ﻟﻴﮕﺎﻧﺪ ﻣﺸﺨﺺ ﮔﺮدﻳﺪ. ﺑﺮرﺳﻲﻫﺎ ﻧﺸﺎن داد ﻛﻪ ﭘﺮوﻟﻴﻦ ﺑﻴﺸﺘﺮﻳﻦ و ﺗﺮﻳﭙﺘﻮﻓﺎن و ﺗﻴﺮوزﻳﻦ ﻛﻤﺘﺮﻳﻦ ﺗﻌﺪاد را دارﻧﺪ. ﻣﺎرﭘﻴﭻ آﻟﻔﺎ ﺣﺪود 50 اﻳﻦ آﻧﺰﻳﻢ را ﺗﺸﻜﻴﻞ ﻣﻲدﻫﺪ، ﻫﻤﭽﻨﻴﻦ ﻣﺸﺨﺺ ﺷﺪ ﻛﻪ اﻳﻦ آﻧﺰﻳﻢ دارای دو داﻣﻨﻪ اﺳﺖ ﻛﻪ ﻳﻜﻲ ﺑﺮای اﺗﺼﺎل ﺑﻪ NAD(P) و دﻳﮕﺮی داﻣﻨﻪ 6 ﻓﺴﻔﻮﮔﻠﻮﻛﻮﻧﺎت اﺳﺖ. ﻣﺤﻞ ﻓﺴﻔﻮرﻳﻠﻪ ﺷﺪن ﻧﻴﺰ ﻣﺸﺨﺺ و ﺑﺎ ﺗﻮﺟﻪ ﺑﻪ ﻋﻤﻠﻜﺮد ﻳﻜﺴﺎن اﻳﻦ آﻧﺰﻳﻢ در ﮔﻴﺎﻫﺎن ﻣﻮرد ﻣﻄﺎﻟﻌﻪ ﻧﻮاﺣﻲ ﺣﻔﺎﻃﺖ ﺷﺪه ﺑﻪ ﻃﻮل 156 اﺳﻴﺪآﻣﻴﻨﻪ ﺑﺮای اﻳﻦ آﻧﺰﻳﻢ ﺗﻌﻴﻴﻦ ﮔﺮدﻳﺪ ﻛﻪ اﻳﻦ ﻧﻮاﺣﻲ ﻧﺸﺎن دﻫﻨﺪه ﺟﺎﻳﮕﺎه ﻓﻌﺎل آﻧﺰﻳﻢ ﻣﻲﺑﺎﺷﻨﺪ. اﻳﻦ ﻋﻮاﻣﻞ ﺑﺮ روی ﺳﺎﺧﺘﺎر ﺳﻪ ﺑﻌﺪی و در ﻧﺘﻴﺠﻪ ﺑﺮ روی ﻋﻤﻠﻜﺮد اﻳﻦ آﻧﺰﻳﻢ ﻣﻮﺛﺮ ﻫﺴﺘﻨﺪ.

The Evaluation of Three Dimensional Structure of Delta- Pyrroline-5-Carboxylate Reductase Enzyme in Plants

Abstract
The evaluation of 3D structure and factors influencing on it can offer better perception from the function of protein. P5CR enzyme catalysis end step of biosynthesis of proline pathway. Proline is a osmolite that modulate osmotic presser in cells under abiotic stress. For this purpose, capture 3D Structure and amino acid sequence for Zea mays, Arabidopsis thaliana, Glycine hispidia and Pisuma sativum from ExPASy and NCBI. with use of bioinformatics tools accomplish evaluation on amino acid sequence and 3D structure, secondary structures, location secondary Structures and type and amount of amino acid in polypeptide chain, location of phosphorylation and ligand binding site. These factors are effective on 3D Structure. The surveys indicate that proline is maximum number of amino acid sequence and tryptophan and tyrosine is in minimum number. Alfa helix constitutes approximately 50% of structure of this enzyme, also this enzyme have two domain that one of them is bind to NAD(P) and other is 6-phosphogloconate. The location of phosphorylation is determined and since this enzyme have same function in these plants, conservation region with 156aa determined for it, these region shows active site of enzyme. These factors are effective on 3D structure and therefore they have an impact on function of enzyme.
