چکیده
ﭼﻐﻨﺪرﻗﻨﺪ ﻳﻜﻲ از ﮔﻴﺎﻫﺎن ﻣﻴﺰﺑﺎن ﮔﻮﻧﻪﻫﺎی ﻣﺨﺘﻠﻒ ﻧﻤﺎﺗﺪ ﻣﻮﻟﺪ ﮔﺮه رﻳﺸﻪ اﺳﺖ. ﺟﻬﺖ ارزﻳﺎﺑﻲ ﻣﻘﺎوﻣﺖ ﭼﻐﻨﺪرﻗﻨﺪ اﺑﺘﺪا ﺗﺄﺛﻴﺮ ﺷﺶ ﺳﻄﺢ زادﻣﺎﻳﻪ 250+250، 500، 250+500، 750، 500+500 و 1000 ﻻرو ﺳﻦ دوم Meloidogyne javanica روی رﻗﻢ ﺣﺴﺎس ﺟﻠﮕﻪ ﺑﺮرﺳﻲ ﺷﺪ. ﺗﻌﺪاد ﮔﺎل ﺗﺸﻜﻴﻞ ﺷﺪه روی رﻳﺸﻪ ﻣﻼک ﻣﻘﺎﻳﺴﻪ ﺗﻴﻤﺎرﻫﺎ ﺑﻮد. ﻧﺘﺎﻳﺞ ﻧﺸﺎن داد ﻛﻪ ﺗﻤﺎﻣﻲ ﺳﻄﻮح زادﻣﺎﻳﻪ ﻗﺎدر ﺑﻪ ارزﻳﺎﺑﻲ ﻣﻘﺎوﻣﺖ ﺑﻮﺗﻪﻫﺎی ﭼﻐﻨﺪرﻗﻨﺪ ﻣﻲﺑﺎﺷﺪ و ﮔﺎﻟﻬﺎی ﺣﺎﺻﻞ از ﻣﺎﻳﻪزﻧﻲ ﻃﻲ ﻳﻚ ﻳﺎ دو ﻧﻮﺑﺖ ﻓﺎﻗﺪ اﺧﺘﻼف آﻣﺎری ﺑﻮدﻧﺪ )0.01≥P(. ﻧﺘﺎﻳﺞ ﻣﺎﻳﻪزﻧﻲ ﻧﻪ ژﻧﻮﺗﻴﭗ ﭼﻐﻨﺪرﻗﻨﺪ ﺑﺎ 500 ﻻرو ﺳﻦ دوم ﻧﺸﺎن داد ﻛﻪ ارﻗﺎم ﺟﻠﮕﻪ و ﭘﺎﺋﻮﻟﺘﺎ )رﻗﻢ ﺗﺠﺎری ﻣﻘﺎوم ﺑﻪ ﻧﻤﺎﺗﺪ ﺳﻴﺴﺘﻲ ﭼﻐﻨﺪرﻗﻨﺪ( ﺑﺎ ﻣﻴﺎﻧﮕﻴﻦ ﮔﺎل ﺑﺎﻻی 100 ﻋﺪد ﺟﺰو ﮔﻴﺎﻫﺎن ﺣﺴﺎس ﺑﻮدﻧﺪ. ﺗﻌﺪاد ﻫﻔﺖ ﻓﺎﻣﻴﻞ ﻧﻴﻤﻪﺧﻮاﻫﺮی ﺗﻬﻴﻪ ﺷﺪه از ﺗﻮده SB33 )ﺣﺎﻣﻞ ژن ﻣﻘﺎوﻣﺖ ﺑﻪ ﻧﻤﺎﺗﺪ ﻣﻮﻟﺪ ﮔﺮه رﻳﺸﻪ( ﺑﺎ ﻣﻴﺎﻧﮕﻴﻦ ﺗﻌﺪاد ﮔﺎل ﻛﻤﺘﺮ از ﻳﻚ ﻧﻴﺰ ﺟﺰو ﮔﻴﺎﻫﺎن ﻣﻘﺎوم دﺳﺘﻪﺑﻨﺪی ﺷﺪﻧﺪ. ﻧﺘﺎﻳﺞ ﻛﻠﻲ ﺣﺎﺻﻞ از اﻳﻦ ﺗﺤﻘﻴﻖ ﻧﺸﺎن داد ﻛﻪ 500 ﻻرو ﺳﻦ دوم ﻧﻤﺎﺗﺪ ﻣﻮﻟﺪ ﮔﺮه رﻳﺸﻪ ﻛﻤﺘﺮﻳﻦ ﺳﻄﺢ زادﻣﺎﻳﻪ ﻧﻤﺎﺗﺪ اﺳﺖ ﻛﻪ ﻣﻲﺗﻮان ﺟﻬﺖ ارزﻳﺎﺑﻲ ﻣﻘﺎوﻣﺖ ﭼﻐﻨﺪرﻗﻨﺪ در ﺷﺮاﻳﻂ ﮔﻠﺨﺎﻧﻪ اﺳﺘﻔﺎده ﻛﺮد.

Optimal inoculum levels for sugar beet resistance screening to root-knot nematode under greenhouse condition

Abstract
Sugar beet (Beta vulgaris) is one of the host for different species root-knot nematodes. For sugar beet resistance screening, firstly the effect of different inoculum levels including 250+250, 500, 500+250, 750, 500+500 and 1000 second stage juveniles of Meloidogyne javanica were studied on the susceptible cultivar jolgeh. Treatments compared based on total number of root galls. Results indicated that all inoculum densities are suitable for screening sugar beet resistance against root-knot nematode and gall numbers in two and one times inoculation have not statistical differences (p≥0.01). Response of nine genotypes to inoculation with 500 second-stage juveniles (in one time) showed cultivars Pauletta (commercial resistant variety to cyst nematode) and Jolgeh with the average number of galls more than 100 are highly susceptible plants. The seven Half-Sib Families derived from SB33 population (with root-knot nematode resistance gene) with the mean number of galls less than 10 were classified as a resistant genotypes. The overall results of this study showed that 500 second stage juvenile root-knot nematode is the lowest inoculum level that can be used for sugar beet resistance screening to root-knot nematode in greenhouse condition.
