چکیده
در اﻳﻦ ﻣﻄﺎﻟﻌﻪ ﻇﺮﻓﻴﺖ رﺷﺪ ﺑﺬرﻫﺎی ﻣﺼﻨﻮﻋﻲ آﻓﺘﺎﺑﮕﺮدان در دو ﺑﺴﺘﺮه ﺗﺠﺎری ﺟﻬﺖ ﻛﺎﺷﺖ ﻣﺴﺘﻘﻴﻢ آنﻫﺎ در ﺷﺮاﻳﻂ ﺑﻴﺮون ﺷﻴﺸﻪای ﻣﻮرد ﺑﺮرﺳﻲ ﻗﺮار ﮔﺮﻓﺖ. آزﻣﺎﻳﺶ ﺑﻪ ﺻﻮرت ﻓﺎﻛﺘﻮرﻳﻞ در ﻗﺎﻟﺐ ﻃﺮح ﻛﺎﻣﻼً ﺗﺼﺎدﻓﻲ ﺑﺎ 5 ﺗﻜﺮار اﻧﺠﺎم ﺷﺪ. ﺟﻨﻴﻦﻫﺎی ﺳﻮﻣﺎﺗﻴﻜﻲ ﺣﺎﺻﻞ از ﻛﺎﺷﺖ ﺟﻨﻴﻦﻫﺎی زﻳﮕﻮﺗﻲ ﻧﺎﺑﺎﻟﻎ آﻓﺘﺎﺑﮕﺮدان در ﻣﺤﻴﻂ ﺣﺎوی ﺳﺎﻛﺎرز ﺑﺎﻻ )mg/l 120 و mg/l BA 2 و mg/l NAA 0/25 در ﻣﺮﺣﻠﻪ ﻛﻮﺗﻴﻠﻪدوﻧﻲ در آﻟﮋﻳﻨﺎت ﺳﺪﻳﻢ 3 و ﻛﻠﺮﻳﺪ ﻛﻠﺴﻴﻢ mM 100 ﻛﭙﺴﻮﻟﻪ ﺷﺪﻧﺪ. در ﺗﻬﻴﻪ ژل آﻟﮋﻳﻨﺎت ﺳﺪﻳﻢ از ﺳﻪ ﻣﺎده ﻣﺨﺘﻠﻒ ﺷﺎﻣﻞ آب ﻣﻘﻄﺮ، ﻣﺤﻴﻂ MS و ﺗﻨﻈﻴﻢ ﻛﻨﻨﺪهﻫﺎی رﺷﺪ اﺳﺘﻔﺎده ﺷﺪ. ﺑﺬرﻫﺎی ﻣﺼﻨﻮﻋﻲ ﺣﺎﺻﻞ ﻣﺴﺘﻘﻴﻤﺎً در دو ﻧﻮع ﺧﺎک ﺷﺎﻣﻞ ﻣﺨﻠﻮط ﻛﻮﻛﻮﭘﻴﺖ و ﭘﺮﻟﻴﺖ و ﻣﺨﻠﻮط ﻛﻮﻛﻮﭘﻴﺖ، ﭘﺮﻟﻴﺖ و ﭘﻴﺖ ﻣﺎس 1:1:1 ﻛﺸﺖ ﺷﺪﻧﺪ. ﻣﻴﺰان ﺗﺒﺪﻳﻞ ﺟﻨﻴﻦﻫﺎی ﺳﻮﻣﺎﺗﻴﻜﻲ ﻛﭙﺴﻮﻟﻪ ﺷﺪه ﺑﻪ ﮔﻴﺎﻫﭽﻪ و رﺷﺪ ﮔﻴﺎﻫﭽﻪﻫﺎی ﺣﺎﺻﻞ در ﺷﺮاﻳﻂ ﺑﻴﺮون ﺷﻴﺸﻪای ﺑﻴﺸﺘﺮ ﺗﺤﺖ ﺗﺄﺛﻴﺮ ﻧﻮع ﻣﺎﺗﺮﻳﻜﺲ آﻟﮋﻳﻨﺎت ﺳﺪﻳﻢ ﻗﺮار ﮔﺮﻓﺖ. در ﺷﺮاﻳﻂ ﺑﻴﺮون ﺷﻴﺸﻪای ﺣﻀﻮر ﻣﻮاد ﻏﺬاﻳﻲ و ﺗﻨﻈﻴﻢ ﻛﻨﻨﺪهﻫﺎی رﺷﺪ در ﻣﺎﺗﺮﻳﻜﺲ ﻛﭙﺴﻮﻟﻪ ﻛﻨﻨﺪه ﺟﻬﺖ ﺣﻤﺎﻳﺖ از ﺟﻮاﻧﻪزﻧﻲ و رﺷﺪ رﻳﺰﻧﻤﻮﻧﻪﻫﺎی ﻛﭙﺴﻮﻟﻪ ﺷﺪه ﺿﺮوری اﺳﺖ.

Evaluation in vivo regeneration of encapsulated somatic embryos of sunflower (Helianthus annuus L.)

A study was set up to determine the regrowth of synthetic seeds of sunflower in two commercial substrates for direct culture them under ex vitro condition. Expriment carried out as factorial randomaized complete block desingn with five replication. Somatic embryos derived from the culture of sunflowerʼs immature zygotic embryos on the medium whit high sucrose (120 g/l) and 2 mg/l BA and 0.25 mg/l NAA in cotyledony stage encapsulated using %3 sodium alginate and 100 mM CaCl2.2H2O. Alginate matrix supplement with three different matrixes include distilled water, liquid MS medium and plant growth regulators. Synthetic seeds sown directly in autoclaved mixture of cocopeat, perlite (1:1) or cocopeat, perlite and peat moss (1:1:1). Conversion into complete plantlets of encapsulated somatic embryos and plantlets regrowth was affected strongly by the composition of calcium alginate beads. Under ex vitro condition presence of MS nutrients and plant growth regulator in the alginate matrix was necessary.
