چکیده
یوﻧﺠﻪ Medicago sativa ﮔﻴﺎﻫﻲ اﺳﺖ ﭼﻨﺪ ﺳﺎﻟﻪ ﻛﻪ ﭘﺮاﻛﻨﺶ وﺳﻴﻌﻲ در اﻳﺮان دارد. ﺑﻪ دﻟﻴﻞ وﺟﻮد ﺟﻤﻌﻴﺖﻫﺎی ﺑﺴﻴﺎر ﮔﺴﺘﺮده و ﻗﺪﻣﺖ اﻳﻦ ﮔﻮﻧـﻪ در ﻛﺸـﻮر و ﺟﻠـﻮﮔ ﻴﺮی از ﻓﺮﺳـﺎ ﻳﺶ ژﻧﺘﻴﻜـ ﻲ آن و ﺣﻔـﻆ اﺻـﺎﻟﺖ ﺑـﺬر ، اﺳـﺘﻔﺎده از ﺗﻜﻨﻮﻟـﻮژ ی ﻓﺮاﺳـﺮد (Cryopreservation) ﺑﺮای ﻧﮕﻬﺪاری ﺑﻠﻨﺪ ﻣﺪت ﺑﺬور اﻳﻦ ﮔﻮﻧﻪ اﻫﻤﻴﺖ وﻳﮋهای دارد. ﺟﻬﺖ ﺑﺮرﺳﻲ رﺷﺪ ﺑﺬور ﻓﺮاﺳـﺮد ی در ﺷﺮاﻳﻂ آزﻣﺎﻳﺸﮕﺎه، ﺑﺬر اﻳﻦ ﮔﻮﻧﻪ از ﻣﺰرﻋﻪ ﺗﺤﻘﻴﻘﺎﺗﻲ ﻫﻤﺪان ﺟﻤﻊ آوری ﺷﺪ. ﺑﻪ ﻣﻨﻈﻮر ﺗﻌﻴﻴﻦ ﺑﻬﺘﺮﻳﻦ ﭘـﻴﺶ ﺗﻴﻤـﺎر ﺟﻬـﺖ ذﺧﻴـﺮه ﺳﺎزی ﺑﺬر در ﻓﺮاﺳﺮد ، ﺳﻪ ﭘﻴﺶ ﺗﻴﻤﺎر ﺷﺎﻣﻞDesiccation, PVS2 و30% Glycerol ﻫﻤﺮاه ﺑﺎ ﺷﺎﻫﺪ ﻣﻮرد آزﻣـﺎﻳﺶ ﻗـﺮار ﮔﺮﻓﺖ. ﺑﺬور ﺗﻴﻤﺎر ﺷﺪه ﺑﻪ ﻣﺪت ﻳﻚﻫﻔﺘﻪ، ﻳﻚﻣﺎه، ﻳﻜﺴﺎل و 26 ﻣﺎه درون ازت ﻣﺎﻳﻊ ﻧﮕﻬﺪاری ﺷـﺪﻧﺪ. ﺑـﺬور ﭘـﺲ از ﺧـﺮوج از ﺑﺮودت 196- درﺟﻪ ﺳﺎﻧﺘﻴﮕﺮاد ، در ﺷﺮاﻳﻂ آزﻣﺎﻳﺸﮕﺎه ﻣﻮرد ﺑﺮرﺳﻲ ﻗﺮار ﮔﺮﻓﺘﻨﺪ و ﺗﻌﺪادی از ﺻـﻔﺎت ﻣـﺮﺗﺒﻂ ﺑـﺎ ﺟﻮاﻧـﻪ زﻧـ ﻲ و اﺳﺘﻘﺮار آﻧﻬﺎ ﺑﺮرﺳﻲ ﮔﺮدﻳﺪ. ﻧﺘﺎﻳﺞ ﻧﺸﺎن داد ﻛﻪ ﺑﺬر Medicago sativa از ﻫﻔﺘﻪ آﻏﺎزﻳﻦ آزﻣﺎﻳﺶ ﺗﺎ 26 ﻣـﺎه ﻗﺎﺑـﻞ ﻧﮕﻬـﺪاری در ﺷﺮاﻳﻂ ﻓﺮاﺳﺮد ﻳﺎ 196 درﺟﺔ ﺳﺎﻧﺘﻴﮕﺮاد ﻣﻲﺑﺎﺷﺪ. در ﺑﺮرﺳﻲ اﺛﺮ ﭘﻴﺶ ﺗﻴﻤﺎرﻫﺎ در ﺣﻔﺎﻇﺖ از ﺑﺬر در ﺷـﺮاﻳﻂ ﻓﺮاﺳـﺮد، ﻣﺸـﺨﺺ ﮔﺮدﻳﺪ ﺗﻔﺎوت ﻣﻌﻨﻲ داری ﺑﻴﻦ ﭘﻴﺶ ﺗﻴﻤﺎر ﻫـﺎی Desiccation ،30%GlycerolوPVS2 ﺑـﺮای اﻏﻠـﺐ ﺻـﻔﺎت ﻣﺸـﺎﻫﺪه ﻣﻲﺷﻮد و ﺑﻬﺘﺮﻳﻦ ﭘﻴﺶ ﺗﻴﻤﺎر Desiccation ﻣﻲﺑﺎﺷﺪ.

Investigation on the possibility of long-term conservation of alfalfa seeds under cryogenic conditions

Alfalfa (Medicago sativa L.) is a perennial forage crop and cultivated in most parts of Iran. Regarding the long-term history of alfalfa cultivation in the country, a numerous populations and high levels of genetic variation observed in the species. In order to maintain genetic diversity, seed genetic characteristics and to prevent genetic erosion, long-term preservation of the seeds via cryopreservation is the most important concerning issue. Seeds of the alfalfa was harvested from Agricultural Research Station of Hamadan and subjected to cryopreservation experiments. In order to achieve suitable pre-cryopreservation treatment, three pre-cryopreservation treatments including 30%Glycerol, PVS2 and Desiccation (including Control) was employed. The treated seeds (except Control) were incubated under liquid nitrogen (LN) conditions or -196°C for 1 week, 1 month, 12 months and 26 months, respectively. The seeds were removed from LN and subjected to heat shock. Some of the seed attributes such as seed germination and establishment were investigated under laboratory conditions. The results showed that Medicago sativa seeds can be long-term preserved under cryogenic (-196°C) conditions. The results also revealed that there are significant differences between pre-cryopreservation treatments including 30%Glycerol, PVS2 and Desiccation for most of the attributes. In this regard, the Desiccation showed excellent cryoprotectant ability compared to those of 30%Glycerol and PVS2.
