چکیده
ﺑﺎدرﻧﺠﺒﻮﻳﻪ ﻳﻜﻲ از ﻗﺪﻳﻤﻲﺗﺮﻳﻦ ﮔﻴﺎﻫﺎن داروﻳﻲ اﺳﺖ ﻛﻪ از زﻣﺎنﻫﺎی دور ﺟﻬﺖ درﻣﺎن ﺑﻴﻤﺎرﻳﻬﺎ اﺳﺘﻔﺎده ﻣﻲﺷﻮد. ﻣﺘﻴﻞﺟﺎﺳﻤﻮﻧﺎت ﻧﻘﺶ ﻣﻬﻤﻲ در رﺷﺪ و ﻧﻤﻮ ﮔﻴﺎه و ﭘﺎﺳﺦ ﺑﻪ ﺗﻨﺶﻫﺎی ﻣﺤﻴﻄﻲ دارد. اﻳﻦ آزﻣﺎﻳﺶ ﺑﻪ ﺻﻮرت ﻓﺎﻛﺘﻮرﻳﻞ و ﺑﺮ ﭘﺎﻳﻪ ﻃﺮح ﻛﺎﻣﻼً ﺗﺼﺎدﻓﻲ اﻧﺠﺎم ﺷﺪ. در اﻳﻦ ﻣﻄﺎﻟﻌﻪ اﺛﺮات ﺳﻄﻮح ﻣﺨﺘﻠﻒ ﻣﺘﻴﻞﺟﺎﺳﻤﻮﻧﺎت، ﺳﺎﻟﻴﺴﻴﻠﻴﻚ اﺳﻴﺪ ﺻﻔﺮ، 0/1، 0/3، 0/5 و ﻳﻚ ﻣﻴﻠﻲﻣﻮﻻر( و ﻣﺤﻴﻂﻫﺎی ﻛﺸﺖ )B5 ،1/2MS ،MS و 1/2B5 ﻣﻮرد ﺑﺮرﺳﻲ ﻗﺮار ﮔﺮﻓﺖ و ﻫﻔﺖ روز ﭘﺲ از ﻣﺤﻠﻮلﭘﺎﺷﻲ ﺑﺮﮔﻲ ﺳﻄﻮح ﻣﺨﺘﻠﻒ ﺗﻴﻤﺎرﻫﺎی ﻳﺎد ﺷﺪه ﺗﻌﺪاد ﺷﺎﺧﺴﺎره، ﻃﻮل ﺑﻠﻨﺪﺗﺮﻳﻦ ﺷﺎﺧﺴﺎره، ﺗﻌﺪاد ﮔﺮه در ﺑﻠﻨﺪﺗﺮﻳﻦ ﺷﺎﺧﺴﺎره، وزن ﺗﺮ و ﺧﺸﻚ ﮔﻴﺎﻫﭽﻪ اﻧﺪازهﮔﻴﺮی ﺷﺪ. ﺑﻴﺸﺘﺮﻳﻦ ﺗﻌﺪاد ﺷﺎﺧﺴﺎره در ﻣﺤﻴﻂ ﻛﺸﺖ MS و در ﻏﻠﻈﺖ ﻳﻚ ﻣﻴﻠﻲﻣﻮﻻر ﻣﺘﻴﻞﺟﺎﺳﻤﻮﻧﺎت و ﺳﻄﻮح 0/3 و ﻳﻚ ﻣﻴﻠﻲﻣﻮﻻر ﺳﺎﻟﻴﺴﻴﻠﻴﻚ اﺳﻴﺪ ﻣﺸﺎﻫﺪه ﺷﺪ. ﻛﻤﺘﺮﻳﻦ ﻃﻮل ﺷﺎﺧﺴﺎره در ﻣﺤﻴﻂ ﻛﺸﺖ MS در ﻏﻠﻈﺖ 0/1 ﻣﻴﻠﻲﻣﻮﻻر ﻣﺘﻴﻞﺟﺎﺳﻤﻮﻧﺎت و ﺑﻴﺸﺘﺮﻳﻦ ﻃﻮل ﺷﺎﺧﺴﺎره ﻧﻴﺰ در ﻏﻠﻈﺖ 0/1 ﻣﻴﻠﻲﻣﻮﻻر ﺳﺎﻟﻴﺴﻴﻠﻴﻚ اﺳﻴﺪ ﻣﺸﺎﻫﺪه ﺷﺪ. ﺑﻴﺸﺘﺮﻳﻦ ﺗﻌﺪاد ﮔﺮه ﻧﻴﺰ در ﻣﺤﻴﻂ ﻛﺸﺖ MS و ﻏﻠﻈﺖ 0/1 ﻣﻴﻠﻲﻣﻮﻻر ﺳﺎﻟﻴﺴﻴﻠﻴﻚ اﺳﻴﺪ ﻣﺸﺎﻫﺪه ﺷﺪ. ﻫﻤﭽﻨﻴﻦ در ﻣﺤﻴﻂ ﻛﺸﺖ MS و ﻏﻠﻈﺖﻫﺎی ﻳﻚ ﻣﻴﻠﻲﻣﻮﻻر ﻣﺘﻴﻞﺟﺎﺳﻤﻮﻧﺎت و 0/1 ﻣﻴﻠﻲﻣﻮﻻر ﺳﺎﻟﻴﺴﻴﻠﻴﻚ اﺳﻴﺪ ﺑﻴﺸﺘﺮﻳﻦ وزن ﺗﺮ و ﺧﺸﻚ ﮔﻴﺎﻫﭽﻪ وﺟﻮد داﺷﺖ. در ﻣﺠﻤﻮع ﻣﺤﻴﻂ ﻛﺸﺖ MS در ﻣﻘﺎﻳﺴﻪ ﺑﺎ ﻣﺤﻴﻂﻫﺎی ﻛﺸﺖ )B5 ،1/2MS و 1/2B5 ﺑﺮﺗﺮ ﺑﻮد.

Effect of different levels of foliar spraying of salicylic acid, methyl jasmonate and medium on shoot growth characteristics of lemon balm to in vitro culture

Abstract
Lemon balm is one of the oldest medicinal plants were common to treatment of disease. Methyl jasmonate plays an important role in Plant growth and response to environmental stress. This experiment was conducted based on completely randomized design. In this study effects of different levels of methyl jasmonate, salicylic acid (0, 0.1, 0.3, 0.5 and 1 mM) and media culture (MS, 1/2MS, B5 and 1/2 B5) in seven days after foliar spraying different levels of treatments was investigated number of shoots, the highest shoot length, number of nodes in the longest shoots, dry and fresh weight of plantlate. The highest number of shoots were observed on MS medium, especially in 1mM concentration of methyl jasmonate and levels of 0.3 and 1mM salicylic acid. The lowest and highest shoot length on MS medium were observed in 0.1 mM methyl jasmonate and 0.1 mM salicylic acid.The highest number of nodes were observed in 0.1 mM salicylic acid. In this research MS medium better than compared with other media culture (1/2MS, B5 and 1/2B5).
