چکیده
ﻫﻤﻴﺸﻚ، ﮔﻴﺎﻫﻲ اﺳﺖ ﺑﺎ ﻧﺎم ﻋﻠﻤﻲDanae racemosa . درﺧﺘﭽﻪ ای زﻳﺒﺎ و ﻫﻤﻴﺸﻪ ﺳﺒﺰ از ﺗﻴﺮه ﻻﻟﻪ1، دارای رﻳﺰوم و ﺷﺎﺧﻪ ﻫﺎی ﺑﺮﮔﻲ (cladodes) اﺳﺖ. اﻳﻦ ﮔﻴﺎه ﺑﻮﻣﻲ اﻳﺮان ﺑﻮده و ﺑﻪ دﻟﻴﻞ رﺷﺪ آﻫﺴﺘﻪ در ﻃﺒﻴﻌﺖ و ﺑﺮداﺷﺖ ﻫﺎی ﺑﻲ روﻳﻪ از آن ﺑﺮای ﮔﻞ آراﻳﻲ،درﻣﻌﺮض ﺧﻄﺮ اﻧﻘﺮاض ﻗﺮار دارد. ﻫﻤﻴﺸﻚ ﮔﻴﺎﻫﻲ اﺳﺖ زﻳﻨﺘﻲ و داروﻳﻲ ﻛﻪ دارای ﺧﻮاص آﻧﺘﻲ اﻛﺴﻴﺪاﻧﻲ و ﺿﺪ اﻧﻌﻘﺎد ﺧﻮن ﺑﻮده و از آن در ﻃﺐ ﺳﻨﺘﻲ و داروﺳﺎزی اﺳﺘﻔﺎده ﻣﻲ ﺷﻮد. ﺟﻮاﻧﻪ زﻧﻲ ﺑﺬرﻫﺎی ﻫﻤﻴﺸﻚ در ﻃﺒﻴﻌﺖ، ﺑﻪ دﻟﻴﻞ ﺧﻔﺘﮕﻲ، ﺣﺪود ﻳﻚ ﺳﺎل ﺑﻪ ﻃﻮل ﻣﻲ اﻧﺠﺎﻣﺪ و ﻇﻬﻮر اوﻟﻴﻦ و دوﻣﻴﻦ ﺑﺮگ آن ﺑﻪ ﺗﺮﺗﻴﺐ در ﺳﺎل ﻫﺎی دوم و ﺳﻮم از ﻛﺎﺷﺖ ﺑﺬر رخ ﻣﻲ دﻫﺪ. ﻫﺪف از اﻳﻦ ﭘﮋوﻫﺶ، ﺑﺮﻃﺮف ﻛﺮدن ﺧﻔﺘﮕﻲ ﺑﺬر و اﻧﺘﺨﺎب ﺗﻴﻤﺎر ﻣﻨﺎﺳﺐ ﺟﻬﺖ ﺟﻮاﻧﻪ زﻧﻲ ﺳﺮﻳﻊ آن در ﺷﺮاﻳﻂ درون ﺷﻴﺸﻪ (in vitro) اﺳﺖ. اﻳﻦ ﺗﺤﻘﻴﻖ ﺑﺮ روی ﺑﺬرﻫﺎی ﻧﺎﺑﺎﻟﻎ و ﻧﻴﻤﻪ ﺑﺎﻟﻎ اﻳﻦ ﮔﻴﺎه اﻧﺠﺎم ﺷﺪ و ﺗﺎﺛﻴﺮ ﺗﻴﻤﺎرﻫﺎی ﻣﺨﺘﻠﻒ از ﻗﺒﻴﻞ ﺳﺮﻣﺎ، ﺟﻴﺒﺮﻟﻴﻚ اﺳﻴﺪ و ﺗﺎرﻳﻜﻲ ﺑﺮ ﺟﻮاﻧﻪ زﻧﻲ ﺑﺬرﻫﺎ در ﺷﺮاﻳﻂ درون ﺷﻴﺸﻪ، آزﻣﺎﻳﺶ ﮔﺮدﻳﺪ. ﻧﺘﺎﻳﺞ ﺣﺎﺻﻞ ﻧﺸﺎن داد ﻛﻪ ﺳﺮﻣﺎدﻫﻲ ﺑﺬرﻫﺎ )دﻣﺎی 3-4درﺟﻪ(، ﺑﻪ ﻣﺪت 10-14 روز و ﺳﭙﺲ ﻗﺮاردادن ﺑﺬرﻫﺎ در ﻣﺤﻴﻂ ﻛﺸﺖ ﻣﻮراﺷﻴﻚ و اﺳﻜﻮگ و ﺗﻴﻤﺎردﻫﻲ ﺑﺎ ﺟﻴﺒﺮﻟﻴﻚ اﺳﻴﺪ و اﻋﻤﺎل ﺷﺮاﻳﻂ ﺗﺎرﻳﻚ در اﺗﺎق ﻛﺸﺖ، ﺑﺮای ﺑﺮﻃﺮف ﻛﺮدن ﺧﻔﺘﮕﻲ ﺑﺬر ﮔﻴﺎه ﻫﻤﻴﺸﻚ ﻣﻨﺎﺳﺐ اﺳﺖ

Breaking of seed dormancy of Danae racemosa in vitro

Abstract
Danae racemosa is a beautiful and evergreen shrub of Liliaceae family which has rhizome and cladodes. This plant is native of Iran and is endangered due to its slow-growing in natural environment and abundant harvesting for flower arrangements. It has also medicinal properties such as antioxidant and antithrombic uses in traditional medicine and pharmacy. Because of seed dormancy, seed germination of Danae racemosa, takes about a year, and the emergence of the first and second leaf occurs in the second and third year of seed culture. The purpose of this study is introducing a method for rapid proliferation of Danae racemosa via in vitro seed culture. Experiments were performed on immature and semi-mature seed so that it was studied the effect of different treatments such as cold, dark and gibberellic acid on in vitro seed germination. The results showed that seed treatment with cold stratification(3-4degrees C) for 10-14 days and followed by seed culture in Murashige and skoog medium supplemented with gibberellic acid in dark condition, is suitable for breaking of seed dormancy of Danae racemosa.
