چکیده
اﺳﺘﻮﻳﻮزاﻳﺪ، ﻳﻜﻲ از ﻣﻬﻢﺗﺮﻳﻦ دیﺗﺮﭘﻦ ﮔﻠﻴﻜﻮزﻳﺪﻫﺎی اﺳﺘﺨﺮاج ﺷﺪه از ﺑﺮگﻫﺎی اﺳﺘﻮﻳﺎ Stevia rebaudiana Bert.)( ﻣﻲﺑﺎﺷﺪ ﻛﻪ ﻋﻼوه ﺑﺮ ﺧﺎﺻﻴﺖ ﺷﻴﺮﻳﻦﻛﻨﻨﺪﮔﻲ ﭼﻨﺪﻳﻦ ﺑﺮاﺑﺮی ﻧﺴﺒﺖ ﺑﻪ ﺳﺎﻛﺎرز، ﺑﻪ ﺳﺒﺐ ﺑﻲاﺛﺮ ﺑﻮدن ﺑﺮ ﻗﻨﺪ ﺧﻮن ﺑﻴﻤﺎران ﻣﺒﺘﻼ ﺑﻪ دﻳﺎﺑﺖ، در ﻃﻲ ﺳﺎلﻫﺎی اﺧﻴﺮ ﻣﻮرد ﺗﻮﺟﻪ ﻗﺮار ﮔﺮﻓﺘﻪ اﺳﺖ. اﻣﺮوزه اﺳﺘﻔﺎده از ﻣﺤﺮکﻫﺎی ﻏﻴﺮ زﻳﺴﺘﻲ ﻧﻈﻴﺮ ﻣﺘﻴﻞ ﺟﺎﺳﻤﻮﻧﺎت و ﺳﺎﻟﻴﺴﻴﻠﻴﻚ اﺳﻴﺪ، در ﺟﻬﺖ اﻓﺰاﻳﺶ ﺗﻮﻟﻴﺪ ﻣﺘﺎﺑﻮﻟﻴﺖﻫﺎی ﺛﺎﻧﻮﻳﻪ در ﻛﺸﺖ ﺑﺎﻓﺖ ﮔﻴﺎﻫﺎن داروﻳﻲ ﻣﻮرد ﺑﺮرﺳﻲ ﻗﺮار ﮔﺮﻓﺘﻪ اﺳﺖ. اﻳﻦ آزﻣﺎﻳﺶ ﺟﻬﺖ ﺳﻨﺠﺶ اﺛﺮ ﻣﺘﻴﻞ ﺟﺎﺳﻤﻮﻧﺎت و ﺳﺎﻟﻴﺴﻴﻠﻴﻚ اﺳﻴﺪ ﺑﺮ ﻣﺤﺘﻮای اﺳﺘﻮﻳﻮزاﻳﺪ اﺳﺘﻮﻳﺎ در ﺷﺮاﻳﻂ درون ﺷﻴﺸﻪای ﺑﻪ ﺻﻮرت ﻓﺎﻛﺘﻮرﻳﻞ ﺑﺮ ﭘﺎﻳﻪ ﻃﺮح ﻛﺎﻣﻼ ﺗﺼﺎدﻓﻲ ﺑﺎ دو ﺗﻜﺮار اﻧﺠﺎم ﺷﺪ. از ﺟﻮاﻧﻪﻫﺎی ﺟﺎﻧﺒﻲ و ﺳﺮﺷﺎﺧﻪﻫﺎ ﺑﻪ ﻋﻨﻮان رﻳﺰﻧﻤﻮﻧﻪ ﺟﻬﺖ ﻛﺸﺖ درون ﺷﻴﺸﻪای در ﻣﺤﻴﻂ ﻛﺸﺖ ﻣﻮراﺷﻴﮓ و اﺳﻜﻮگ )MS1(اﺳﺘﻔﺎده ﮔﺮدﻳﺪ. ﭘﺲ از واﻛﺸﺖ رﻳﺰﻧﻤﻮﻧﻪﻫﺎ در ﻣﺤﻴﻂ ﻛﺸﺖ MS و ﮔﺬﺷﺖ دوره زﻣﺎﻧﻲ 30 روزه، ﺗﻴﻤﺎر ﺑﺎ ﺳﻄﻮح ﻣﺨﺘﻠﻒ ﻣﺘﻴﻞ ﺟﺎﺳﻤﻮﻧﺎت و ﺳﺎﻟﻴﺴﻴﻠﻴﻚ اﺳﻴﺪ )ﺻﻔﺮ، 0/5 و ﻳﻚ ﻣﻴﻠﻲﻣﻮﻻر( ﺑﻪ ﺻﻮرت ﻣﺤﻠﻮلﭘﺎﺷﻲ ﺑﺮﮔﻲ اﻧﺠﺎم ﺷﺪ. ﭘﺲ از ﮔﺬﺷﺖ 14 روز و اﺳﺘﺨﺮاج ﻋﺼﺎره ﻣﺘﺎﻧﻮﻟﻲ از اﻧﺪامﻫﺎی ﻫﻮاﻳﻲ ﮔﻴﺎﻫﭽﻪﻫﺎ، اﻧﺪازهﮔﻴﺮی ﻣﺤﺘﻮای اﺳﺘﻮﻳﻮزاﻳﺪ ﺑﻪ روش ﻛﺮوﻣﺎﺗﻮﮔﺮاﻓﻲ ﻣﺎﻳﻊ ﺑﺎ ﻛﺎراﻳﻲ ﺑﺎﻻ اﻧﺠﺎم ﺷﺪ. ﻧﺘﺎﻳﺞ ﻧﺸﺎن داد ﻛﻪ ﻣﺤﺘﻮای اﺳﺘﻮﻳﻮزاﻳﺪ در ﮔﻴﺎﻫﭽﻪﻫﺎی ﻣﺤﻠﻮلﭘﺎﺷﻲ ﺷﺪه ﺑﺎ ﻣﺘﻴﻞ ﺟﺎﺳﻤﻮﻧﺎت و ﺳﺎﻟﻴﺴﻴﻠﻴﻚ اﺳﻴﺪ در ﻣﻘﺎﻳﺴﻪ ﺑﺎ ﺗﻴﻤﺎر ﺷﺎﻫﺪ اﻓﺰاﻳﺶ ﻣﻌﻨﻲداری ﻳﺎﻓﺖ. ﻫﻤﭽﻨﻴﻦ ﻣﺤﻠﻮلﭘﺎﺷﻲ ﺑﺎ ﻣﺘﻴﻞ ﺟﺎﺳﻤﻮﻧﺎت اﺛﺮ ﺑﻴﺸﺘﺮی ﺑﺮ ﻣﺤﺘﻮای اﺳﺘﻮﻳﻮزاﻳﺪ در ﻣﻘﺎﻳﺴﻪ ﺑﺎ ﺳﺎﻟﻴﺴﻴﻠﻴﻚ اﺳﻴﺪ داﺷﺖ.

Effects of methyl jasmonate and salicylic acid on stevioside production in in vitro culture of Stevia rebaudiana Bertoni

Stevioside, an ent-kaurene type of diterpenoid glycoside, is a natural sweetener extracted from leaves of stevia rebaudiana Bertoni a perennial herb of the asteraceae family that can be used as substitute of artificial sweeteners for diabetic patients. Elicitors could be used as enhancers of plant secondary metabolites synthesis and could play an important role in biosynthetic pathways to production of commercially important compounds. This investigation aimed to study the improving effect of methyl jasmonate and salicylic acid foliar spray application (0, 0.5 and 1 mM) on stevioside content of stevia in in-vitro conditions. Shoot tip and nodal segment were used as explants and then were cultured on Murashige and Skoog medium. After 30 days, both of the elicitor’s effects on the production of stevioside were studied. Plantlets were harvested after 14 days and high-performance liquid chromatography was used to assess and quantification of stevioside content. Experiment was carried out as factorial based on completely randomized design (CRD) with two replications. The results showed that plantlets of stevia treated with methyl jasmonate and salicylic acid increased the content of stevioside compared to non-treated plantlets. It was also noticed that foliar spray with methyl jasmonate had higher effect on stevioside content than salicylic acid.
