چکیده
ﻋﻠﻒ ﭼﺎی Hypericum perforatum ﻣﺘﻌﻠﻖ ﺑﻪ ﺧﺎﻧﻮاده Hypericaceae ﻳﻚ ﮔﻴﺎه داروﻳﻲ ارزﺷﻤﻨﺪ اﺳﺖ ﻛﻪ از ﮔﺬﺷﺘﻪﻫﺎی دور ﺑﺮای اﻫﺪاف ﭘﺰﺷﻜﻲ ﻛﺎرﺑﺮد داﺷﺘﻪ و ﻣﻮاد ﻣﺆﺛﺮ آن از ﺟﻤﻠﻪ ﻫﺎﻳﭙﺮﺳﻴﻦ و ﻫﺎﻳﭙﺮﻓﻮرﻳﻦ دارای ﺧﻮاص ﻣﻬﻤﻲ از ﺟﻤﻠﻪ ﺧﺎﺻﻴﺖ ﺿﺪاﻓﺴﺮدﮔﻲ، ﺿﺪاﻟﺘﻬﺎﺑﻲ و ﺿﺪ ﺑﺎﻛﺘﺮی ﻣﻲﺑﺎﺷﻨﺪ.اﺧﻴﺮا اﺳﺘﻔﺎده از ﻣﺤﺮکﻫﺎ ﺑﻪ ﻋﻨﻮان ﻋﻮاﻣﻠﻲ ﻛﻪ ﻣﻲﺗﻮاﻧﻨﺪ ﺗﻮﻟﻴﺪ ﺑﻴﺶ از ﻧﻴﺎز ﻣﺘﺎﺑﻮﻟﻴﺖﻫﺎی ﺛﺎﻧﻮﻳﻪ در ﮔﻴﺎه اﻟﻘﺎء ﻛﻨﻨﺪ ﻣﻮرد ﺗﻮﺟﻪ ﻗﺮار ﮔﺮﻓﺘﻪ اﺳﺖ. در اﻳﻦ ﺗﺤﻘﻴﻖ اﺛﺮ ﺳﺎﻟﻴﺴﻴﻠﻴﻚ اﺳﻴﺪ ﺑﺎ ﻏﻠﻈﺖﻫﺎی ﺻﻔﺮ ، 200 ، 400 ، 600 ﻣﻴﻜﺮوﻣﻮﻻر در ﭼﻬﺎر ﻣﺤﻴﻂ ﻛﺸﺖ ﭘﺎﻳﻪ ﺷﺎﻣﻞ: MS و B5 )100 و 50 درﺻﺪ( ﺑﺼﻮرت ﻓﺎﻛﺘﻮرﻳﻞ در ﭘﺎﻳﻪ ﻃﺮح ﻛﺎﻣﻼً ﺗﺼﺎدﻓﻲ ﺑﺎ ﺳﻪ ﺗﻜﺮار ﺑﺮ ﺧﺼﻮﺻﻴﺎت ﻣﻮرﻓﻮﻟﻮژﻳﻚ ﻣﻮرد ﺑﺮرﺳﻲ ﻗﺮار ﮔﺮﻓﺖ. ﻧﺘﺎﻳﺞ اﻳﻦ ﺑﺮرﺳﻲ ﻧﺸﺎن داد ﻛﻪ ﺑﻴﻦ ﻣﺤﻴﻂﻫﺎی ﻛﺸﺖ از ﻧﻈﺮ ﺗﺄﺛﻴﺮ ﺑﺮ ﺗﻌﺪاد و ﻃﻮل ﺷﺎﺧﺴﺎره، ﺗﻌﺪادﮔﺮه، ﺷﺎﺧﺺ ﻛﻠﺮوﻓﻴﻞ ﺑﺮگ اﺧﺘﻼف ﻣﻌﻨﻲدار ﻣﺸﺎﻫﺪه ﺷﺪ. ﺳﺎﻟﻴﺴﻴﻠﻴﻚ اﺳﻴﺪ ﻧﻴﺰ ﺗﺄﺛﻴﺮ ﻣﻌﻨﻲداری ﺑﺮ ﺗﻤﺎم ﺻﻔﺎت ﻣﻮرد ﺑﺮرﺳﻲ ﺑﺠﺰ ﺗﻌﺪاد ﮔﺮه در ﺷﺎﺧﺴﺎره داﺷﺖ. ﻫﻤﭽﻨﻴﻦ ﺑﺎ ﻣﻘﺎﻳﺴﻪ ﻣﻴﺎﻧﮕﻴﻦﻫﺎ، ﺑﻴﺸﺘﺮﻳﻦ ﺗﻌﺪاد ﺷﺎﺧﺴﺎره در ﻣﺤﻴﻂﻫﺎی B5 و 1/2B5 ودر ﻏﻠﻈﺖ400 ﻣﻴﻜﺮوﻣﻮﻻر ﺳﺎﻟﻴﺴﻴﻠﻴﻚ اﺳﻴﺪ ﻣﺸﺎﻫﺪه ﺷﺪ، ﻫﻤﭽﻨﻴﻦ در ﻫﺮ ﭼﻬﺎر ﻣﺤﻴﻂ ﻛﺸﺖ اﻓﺰودن ﻏﻠﻈﺖ ﺳﺎﻟﻴﺴﻴﻠﻴﻚ اﺳﻴﺪ ﺑﻪ ﻣﻴﺰان 400 ﻣﻴﻜﺮوﻣﻮﻻر ﺑﺎﻋﺚ اﻓﺰاﻳﺶ ﺷﺎﺧﺺ ﻛﻠﺮوﻓﻴﻞ ﮔﺮدﻳﺪ. ﻃﻮل ﺷﺎﺧﺴﺎره و ﺗﻌﺪاد ﮔﺮه در ﺑﻠﻨﺪﺗﺮﻳﻦ ﺷﺎﺧﺴﺎره در ﻣﺤﻴﻂ ﻛﺸﺖ MS ﺑﻄﻮر ﻣﺘﻮﺳﻂ ﻧﺴﺒﺖ ﺑﻪ ﻣﺤﻴﻂ ﻛﺸﺖ B5 ﺑﻴﺸﺘﺮ ﺑﻮد.

INFLUENCE OF SALICYLIC ACID ON MORPHOLOGICAL CHARACTERICTICS AND CHLOROPHYLL CONTENT IN IN VITRO CULTURE OF HYPERICUM PERFORATUM

Abstract
hypericum perforatum from hypericaceae family is important medicinal plant and contains active ingredients including hypericin and hyperforin. This plant has anti-bacterial, antidepressants and anti-inflammatary properties. This study was undertaken to investigation of effect of different concentrations of salicylic acid (0, 200, 400, 600 micro molar (µ.M)) and MS and B5 medium concentrations (50 % and 100%). Salicylic acid treatments were applied after 40 days. Experiment was conducted in a completely randomized design with three replications. Teen days after the treatments, morphological characteristic and leaf chlorophyll index was examined. The results indicate significant differences between the culture mediums in the shoot number and shoot length, node number in longest shoot, and chlorophyll content. Salicylic acid had significant effect on all characteristics except node number. The most shoot number was shown in B5 and ½ B5 medium supplemented with 400 µ.M of salicylic acid, in four culture media increasing salicylic acid concentration up to 400 µ.M, led to increase the chlorophyll content. Based on shoot length and node number on longest shoot, MS mediums was better than B5 mediums.
