چکیده
ﻋﻠﻒ ﭼﺎی Hypericum perforatum از ﺧﺎﻧﻮاده Hypericaceae ﺗﺄﺛﻴﺮات ﻓﺎرﻣﺎﻛﻮﻟﻮژﻳﻜﻲ ﻣﺘﻌﺪدی از ﺟﻤﻠﻪ ﺿﺪ اﻓﺴﺮدﮔﻲ و ﺿﺪ اﻟﺘﻬﺎﺑﻲ را داراﺳﺖ. اﻳﻦ ﺗﺤﻘﻴﻖ در راﺳﺘﺎی ارزﻳﺎﺑﻲ ﺗﺎﺛﻴﺮ ﻣﺘﻴﻞ ﺟﺎﺳﻤﻮﻧﺎت ﺑﺎ ﻏﻠﻈﺖ ﻫﺎی ﻣﺨﺘﻠﻒ )ﺻﻔﺮ، 300، 600و 900 ﻣﻴﻜﺮوﻣﻮﻻر( و در ﻣﺤﻴﻂ ﻫﺎی ﻛﺸﺖ (MS , B5 در ﻏﻠﻈﺖ◌ٰ ﻫﺎی 50 و 100 درﺻﺪ در ﺷﺮاﻳﻂ درون ﺷﻴﺸﻪ ای اﻧﺠﺎم ﺷﺪ. ﺑﺬور ﺑﺎ اﺗﺎﻧﻮل 70 درﺻﺪ و ﻫﻴﭙﻮﻛﻠﺮﻳﺪ ﺳﺪﻳﻢ 50 درﺻﺪ اﺳﺘﺮﻳﻞ ﺷﺪ و ﺳﭙﺲ در ﻣﺤﻴﻄﻬﺎی ﻛﺸﺖ ﻣﺨﺘﻠﻒ ﻛﺸﺖ ﮔﺮدﻳﺪ. آزﻣﺎﻳﺶ در ﺷﺮاﻳﻂ ﻃﺮح ﻛﺎﻣﻼً ﺗﺼﺎدﻓﻲ ﺑﺎ ﺳﻪ ﺗﻜﺮار اﻧﺠﺎم ﺷﺪ. ﻣﺘﻴﻞ ﺟﺎﺳﻤﻮﻧﺎت ﺑﺼﻮرت اﺳﭙﺮی ﭘﺲ از 60 روز از رﺷﺪ ﮔﻴﺎﻫﺎن ﺑﺮ روی آن ﻫﺎ ﺗﻴﻤﺎر ﺷﺪ.10روز ﭘﺲ از ﺗﻴﻤﺎر رﺷﺪ رﻳﺸﻪ و ﮔﻴﺎﻫﭽﻪ اﻧﺪازه ﮔﻴﺮی ﺷﺪ. ﻧﺘﻴﺠﻪ ﻧﺸﺎن داد ﻛﻪ ﺑﻴﻦ ﻣﺤﻴﻂ ﻫﺎی ﻛﺸﺖ ﺑﻜﺎر ﺑﺮده ﺷﺪه در ﺻﻔﺎت وزن ﺗﺮ رﻳﺸﻪ و ﮔﻴﺎﻫﭽﻪ ارﺗﺒﺎط ﻣﻌﻨﻲ داری وﺟﻮد داﺷﺖ. ﻏﻠﻈﺖ ﻣﺘﻴﻞ ﺟﺎﺳﻤﻮﻧﺎت ﺗﺎﺛﻴﺮ ﻣﻌﻨﻲ داری ﺑﺮ روی ﺗﻤﺎم ﺻﻔﺎت ﺑﺠﺰ وزن ﺧﺸﻚ ﮔﻴﺎﻫﭽﻪ داﺷﺖ.ﺗﺒﺎط ﻣﻌﻨﻲ داری ﺑﻴﻦ ﻣﺤﻴﻂ ﻫﺎی ﻛﺸﺖ و ﻣﺘﻴﻞ ﺟﺎﺳﻤﻮﻧﺎت در وزن ﺗﺮ رﻳﺸﻪ و ﮔﻴﺎﻫﭽﻪ وﺟﻮد داﺷﺖ. در ﻣﺤﻴﻂ ﻛﺸﺖ B5 ,1/2 وزن ﺧﺸﻚ ﮔﻴﺎﻫﭽﻪ ﻛﻤﺘﺮ از ﻣﺤﻴﻂ ﻛﺸﺖ ﻫﺎی MS ,1/2MS ﺑﻮد. ﻧﺘﺎﻳﺞ ﻧﺸﺎن داد ﭘﺎﺳﺦ رﺷﺪی
Hypericum ﺑﻪ ﻣﺘﻴﻞﺟﺎﺳﻤﻮﻧﺎت در ﻣﺤﻴﻂ ﻛﺸﺖ MS ﺑﻬﺘﺮ از ﻣﺤﻴﻂ ﻛﺸﺖ5 B بود.

EFFECT OF METHYL JASMONATE AND MEDIA CULTURE ON ROOT AND PLANTLET GROWTH IN HYPERICUM PERFORATUM

Abstract
Hypericum perforatum from hypericaceae has several pharmacological effects like anti-depressant and antiinflammatory.
This study aimed to evaluate the effect of methyl jasmonate with various concentration (0,
300, 600, 900 micro molar (μ.M)) and culture Media MS and B5 (50% , 100%) on Hypericum perforatum in
in-vitro culture condition. The seeds sterilized by 70% ethanol and sodium hypochlorite (50%), and then
were cultured in different culture media. Experiment was conducted in a completely randomized design with
three replications. Methyl jasmonate treatments were applied after 40 days. Fourteen days after the
treatments, root and plantlet growth and hypericin concentration were Measured. The results showed that
between culture media based on dry and wet weight of roots and plantlet there was significant difference.
Methyl jasmonate concentrations had significant effect on all of characteristcs except plantlet dry weight.
There was significant interaction between mediums and methyl jasmonate based on wet and dry
weight of root and plantlet and Hypericin concentration. Dry weight of roots in ½ B5 was lower than
others. In B5 and ½ B5, dry weight of plantlet was lower than MS and ½ MS. Our results showed that the
response of Hypericum perforatum to methyl jasmonate in MS medium is better than B5.
