چکیده
اﺳﺘﻮﻳﺎ (Stevia rebaudiana) ﻳﻚ ﮔﻴﺎه داروﻳﻲ از ﺧﺎﻧﻮاده آﺳﺘﺮاﺳﻪ اﺳﺖ ﻛﻪ ﺑﻪ دﻟﻴﻞ ﺧﺎﺻﻴﺖ ﺷﻴﺮﻳﻦ ﻛﻨﻨﺪﮔﻲ ﺑﺎﻻ )ﺣﺪود 200 اﻟﻲ 300 ﻣﺮﺗﺒﻪ ﺷﻴﺮﻳﻦ ﺗﺮ از ﺷﻜﺮ( از اﻫﻤﻴﺖ اﻗﺘﺼﺎدی ﺑﺎﻻﻳﻲ ﺑﺮﺧﻮردار ﻫﺴﺖ. اﻣﺎ ﭘﺎﻳﻴﻦ ﺑﻮدن درﺻﺪ ﺟﻮاﻧﻪزﻧﻲ و ﭘﻮﻛﻲ ﺑﺬر ﻛﺸﺖ وﺳﻴﻊ آن را ﻣﺤﺪود ﻛﺮده اﺳﺖ. ﻛﺸﺖ درون ﺷﻴﺸﻪای ﻳﻚ روش ﻣﻮﺛﺮ ﺑﺮای ﺗﻮﻟﻴﺪ اﻧﺒﻮه اﺳﺘﻮﻳﺎ ﻣﺤﺴﻮب ﻣﻲﺷﻮد. اﻳﻦ ﭘﮋوﻫﺶ ﺑﻪ ﻣﻨﻈﻮر ﻣﻄﺎﻟﻌﻪ ﺳﺎزﮔﺎری ﮔﻴﺎﻫﭽﻪﻫﺎی ﺗﻮﻟﻴﺪ ﺷﺪه اﺳﺘﻮﻳﺎ در ﺷﺮاﻳﻂ درون ﺷﻴﺸﻪای ﺑﻪ ﻣﺤﻴﻂ آزاد و ﮔﻠﺨﺎﻧﻪای اﻧﺠﺎم ﺷﺪ. ﺑﺪﻳﻦ ﻣﻨﻈﻮر آزﻣﺎﻳﺸﻲ ﺑﺎ اﺳﺘﻔﺎده از ﺳﻪ ﻣﺤﻠﻮل آﺑﻴﺎری آب ﺷﻬﺮی، 1/2MS و ﻫﻮﮔﻠﻨﺪ( و ﺷﺶ ﺑﺴﺘﺮ ﻛﺸﺖ (ﺧﺎک زراﻋﻲ، ﻣﺎﺳﻪ، ﭘﺮﻟﻴﺖ، ﺷﻦ، ﺷﻦ، ﭘﺮﻟﻴﺖ+ ﻣﺎﺳﻪ و ﺷﻦ+ ﻣﺎﺳﻪ) اﻧﺠﺎم ﺷﺪ. آزﻣﺎﻳﺶ ﺑﻪ ﺻﻮرت ﻓﺎﻛﺘﻮرﻳﻞ در ﻗﺎﻟﺐ ﻃﺮح ﻛﺎﻣﻼ ﺗﺼﺎدﻓﻲ ﺻﻮرت ﮔﺮﻓﺖ. ﭘﺲ از 4 ﻫﻔﺘﻪ ﺻﻔﺎت درﺻﺪ ﺑﻘﺎ و ﻃﻮل ﮔﻴﺎﻫﭽﻪﻫﺎ ﻣﻮرد ارزﻳﺎﺑﻲ ﻗﺮار ﮔﺮﻓﺘﻨﺪ. ﻧﺘﺎﻳﺞ ﺣﺎﺻﻞ ﻧﺸﺎن داد ﻛﻪ ﺑﻴﺸﺘﺮﻳﻦ درﺻﺪ ﺑﻘﺎ و ﻃﻮل ﮔﻴﺎﻫﭽﻪ از ﺑﺴﺘﺮﻫﺎی ﭘﺮﻟﻴﺖ و ﺷﻦ ﺑﺎ ﻣﺤﻠﻮل ﻏﺬاﻳﻲ ﻫﻮﮔﻠﻨﺪ ﺗﻮﻟﻴﺪ ﺷﺪه اﺳﺖ.

Study of Adaptation In vitro Plants of Stevia (Stevia rebaudiana) to Free Environment

Abstract
Stevia (Stevia rebaudiana) is a medicinal plant belonging to the family of asteraceae. It have great economic, Because this plant have high sweeteners property. At the first step low viability and low seed germination, limit their extensive cultivation. One of the effective methods to produce this plant is in vitro technology that solve the problem of large scale production. The purpose of this investigation is the study of adaptation of in vitro plants to the free environment. For this purpose were used of six culture substrate (crop soil, sand, perlite, gravel, perlite + sand (1:1) and gravel + sand (1:1)) with three irrigation solution (drinking water, 1/2 MS and hoagland solution), That was conducted in a factorial experiment based on randomized complete design with three replications. After 4 weeks length of shoot and survival percentage were evaluated. The results showed that the maximum length of shoot and survival percentage obtained in culture substrate of perlite and gravel with hoagland nutrient solution.
