چکیده
ﺳﺮﻃﺎن ﻳﻜﻲ از ﻋﻠﻞ ﻣﻬﻢ ﻣﻨﺠﺮ ﺑﻪ ﻣﺮگ در ﺳﺮاﺳﺮ ﺟﻬﺎن، ﺑﻮﻳﮋه در ﻛﺸﻮرﻫﺎی در ﺣﺎل ﺗﻮﺳﻌﻪ اﺳﺖ. ﺳﺮﻃﺎن ﺳﻴﻨﻪ ﺷﺎﻳﻊﺗﺮﻳﻦ ﻧﻮع ﺳﺮﻃﺎن در ﻣﻴﺎن زﻧﺎن اﺳﺖ، و ﺳﺎﻻﻧﻪ ﺑﻴﺶ از ﻧﻴﻢ ﻣﻴﻠﻴﻮن زن ﻣﺒﺘﻼ ﺑﻪ اﻳﻦ ﺑﻴﻤﺎری ﻣﻲﺷﻮﻧﺪ. Trastuzumab )ﺑﺎ ﻧﺎم ﺗﺠﺎری Herceptin( ﻳﻚ آﻧﺘﻲﺑﺎدی اﻧﺴﺎﻧﻲ ﺷﺪه ﻣﺸﺘﻖ از DNA ﻧﻮﺗﺮﻛﻴﺐ اﺳﺖ ﻛﻪ در درﻣﺎن ﺳﺮﻃﺎن ﻣﺘﺎﺳﺘﺎﺗﻴﻚ ﺳﻴﻨﻪ ﻣﻮرد اﺳﺘﻔﺎده ﻗﺮار ﻣﻲﮔﻴﺮد. ﻛﺸﺎورزی ﻣﻮﻟﻜﻮﻟﻲ" ﮔﻴﺎﻫﻲ ﺑﻪ ﻋﻨﻮان ﻳﻚ روش اﻣﻴﺪﺑﺨﺶ ﺑﺎ ﻣﺰاﻳﺎی ﻗﺎﺑﻞ ﺗﻮﺟﻪ از ﻧﻈﺮ ﻫﺰﻳﻨﻪ و اﻳﻤﻨﻲ ﻧﺴﺒﺖ ﺑﻪ ﺳﺎﻳﺮ ﺳﻴﺴﺘﻢﻫﺎی ﺑﻴﺎن ﻳﻮﻛﺎرﻳﻮﺗﻲ ﺷﻨﺎﺧﺘﻪ ﺷﺪه اﺳﺖ. در ﺗﺤﻘﻴﻖ ﺣﺎﺿﺮ Agrobacterium tumefaciens ﺳﻮﻳﻪ LBA4404 درﺑﺮدارﻧﺪه ﭘﻼﺳﻤﻴﺪ pBIN-Her2-HLC ﺣﺎوی ژنﻫﺎی رﻣﺰ ﻛﻨﻨﺪهی Trastuzumab و ژن nptII )ﺑﻪ ﻋﻨﻮان ژن ﻧﺸﺎﻧﮕﺮ اﻧﺘﺨﺎﺑﻲ( ﺑﺮای ﺗﺮارﻳﺨﺘﻲ ﮔﻴﺎه ﺗﻮﺗﻮن )Nicotiana tubaccum(
ﻣﻮرد اﺳﺘﻔﺎده ﻗﺮار ﮔﺮﻓﺖ. اﻧﺘﺨﺎب اوﻟﻴﻪ ﺳﻠﻮلﻫﺎی ﺗﺮارﻳﺨﺖ ﺑﺮ روی ﻣﺤﻴﻂ ﺣﺎوی ﻛﺎﻧﺎﻣﺎﻳﺴﻴﻦ )mg/l 150 اﻧﺠﺎم ﮔﺮﻓﺖ. ﺣﻀﻮر ژنﻫﺎی ﻫﺪف در ﮔﻴﺎﻫﭽﻪﻫﺎی ﺗﺮارﻳﺨﺘﻪ اﺣﺘﻤﺎﻟﻲ ﺑﺎ اﺳﺘﻔﺎده از آﻧﺎﻟﻴﺰ PCR ﺗﺎﻳﻴﺪ ﺷﺪ. ﮔﻴﺎﻫﭽﻪﻫﺎی ﺗﺮارﻳﺨﺘﻪ رﻳﺸﻪدار ﺷﺪه ﺑﻪﻃﻮر ﻣﻮﻓﻘﻴﺖآﻣﻴﺰ و ﺑﺎ ﻗﺎﺑﻠﻴﺖ زﻧﺪهﻣﺎﻧﻲ 100 در ﻣﺤﻴﻂ ﮔﻠﺨﺎﻧﻪ ﺳﺎزﮔﺎر ﺷﺪﻧﺪ. آﻧﻬﺎ وﻳﮋﮔﻲﻫﺎی ﻣﻮرﻓﻮﻟﻮژﻳﻜﻲ ﻣﺸﺎﺑﻪ ﺑﺎ ﮔﻴﺎﻫﺎن ﻣﺎدری ﻧﺸﺎن دادﻧﺪ.

Agrobacterium tumefaciens-mediated transformation of tobacco with the genes encoding the monoclonal antibody trastuzumab

Abstract
Cancer is one of the leading causes of death in the world, particularly in developing countries. Breast cancer is the most common cancer among women, annually more than half a million women were diagnosed with breast cancer. Trastuzumab (Herceptin) is a recombinant DNA-derived humanized antibody that is used in the treatment of metastatic breast cancer. Plant-based “molecular farming” has revealed as a promising approach with significant advantages in both cost and safety over other eukaryotic expression systems. In the present study, Agrobacterium tumefaciens strain LBA4404 harboring pBIN-Her2-HLC plasmid containing the genes encoding trastuzumab and the nptII gene (as a selectable marker gene) were used for tobacco plant transformation. Initial selection of transgenic cells was performed on medium containing kanamycin (150 mg/l). The presence of the target genes in putative transformants was confirmed by PCR analyses. The rooted transgenic plantlets were acclimatized successfully in the greenhouse, with a survival rate of 100%. They showed normal morphological characteristics similar to the maternal plant.
