چکیده
ﻫﺘﺮوزﻳﺲ ﺗﻮﺳﻂ ژنﻫﺎ ﭘﺮوﺗﺌﻴﻦﻫﺎﻳﻲ ﻛﻨﺘﺮل ﻣﻲﺷﻮﻧﺪ ﻛﻪ ﺷﻨﺎﺳﺎﻳﻲ اﻳﻦ ژنﻫﺎ و ﭘﺮوﺗﺌﻴﻦﻫﺎ از دﻳﺪﮔﺎه ﻣﻠﻜﻮﻟﻲ در ﭘﺮوژهﻫﺎی اﺻﻼح ﮔﻨﺪم، از اﻫﻤﻴﺖ ﺑﺴﻴﺎری ﺑﺮﺧﻮردار اﺳﺖ. ﺑﻪ ﻣﻨﻈﻮر ﺑﺮرﺳﻲ وﺟﻮد اﺛﺮ ﻫﺘﺮوزﻳﺲ در ﻣﻘﺎوﻣﺖ ﺑﻪ ﺑﻴﻤﺎری ﺳﭙﺘﻮرﻳﻮز ﺑﺮگ ﮔﻨﺪم ارﻗﺎم زاﮔﺮس )ﺣﺴﺎس(، N8118 )ﻣﻘﺎوم( و ﻫﻴﺒﺮﻳﺪ ﺣﺎﺻﻞ از آﻧﻬﺎ در اﻳﺴﺘﮕﺎه ﺗﺤﻘﻴﻘﺎت ﻛﺸﺎورزی ﮔﺮﮔﺎن در ﺳﻪ ﺗﻜﺮار و در ﻗﺎﻟﺐ ﻃﺮح ﺑﻠﻮک ﻛﺎﻣﻞ ﺗﺼﺎدﻓﻲ ﻛﺸﺖ ﮔﺮدﻳﺪﻧﺪ. ﻣﺎﻳﻪ زﻧﻲ ﻣﺼﻨﻮﻋﻲ ارﻗﺎم در ﻳﻚ ﻧﻮﺑﺖ اﻧﺠﺎم ﮔﺮﻓﺖ. ﺟﻬﺖ ﺑﺮرﺳﻲ وﺟﻮد اﺛﺮ ﻫﺘﺮوزﻳﺲ از آﻧﺎﻟﻴﺰ ﭘﺮوﺗﺌﻮم ﺑﺎﻓﺖ ﺑﺮگ ﭘﺮﭼﻢ اﺳﺘﻔﺎده و اﺳﺘﺨﺮاج ﭘﺮوﺗﺌﻴﻦ ﺑﺮ اﺳﺎس روش TCA/Aceton ﺗﻐﻴﻴﺮ ﻳﺎﻓﺘﻪ اﻧﺠﺎم ﺷﺪ. ﻧﺘﺎﻳﺞ ﺣﺎﺻﻞ از آﻧﺎﻟﻴﺰ ژلﻫﺎ ﻗﺒﻞ از ﻣﺮﺣﻠﻪ اﺳﭙﻮر ﭘﺎﺷﻲ ﺗﻌﺪاد 5 ﻟﻜﻪ ﭘﺮوﺗﺌﻴﻨﻲ ﺑﺎ ﺑﻴﺎن اﻓﺘﺮاﻗﻲ ﺑﻴﻦ واﻟﺪﻳﻦ و ﻫﻴﺒﺮﻳﺪ را ﻣﺸﺨﺺ ﻧﻤﻮد. ﻫﻤﭽﻨﻴﻦ ﺣﻀﻮر ﻳﻚ ﻟﻜﻪ ﭘﺮوﺗﺌﻴﻨﻲ ﺑﺮ روی ژل ﻫﻴﺒﺮﻳﺪ و ﻋﺪم ﺣﻀﻮر ﻫﻤﺎن ﻟﻜﻪ ﺑﺮ روی ژلﻫﺎی واﻟﺪﻳﻦ ﺑﻌﺪ از اﺳﭙﻮر ﭘﺎﺷﻲ اﺣﺘﻤﺎل وﺟﻮد ﻫﺘﺮوزﻳﺲ را ﺗﺎﺋﻴﺪ ﻧﻤﻮد. ﻧﺘﺎﻳﺞ ﺣﺎﺻﻞ از اﻳﻦ ﺗﺤﻘﻴﻖ ﻧﺸﺎن ﻣﻲدﻫﺪ درﺻﻮرت اﻓﺰاﻳﺶ اﻃﻼﻋﺎت ﻣﻠﻜﻮﻟﻲ در ﻣﻮرد ﭘﺪﻳﺪه ﻫﺘﺮوزﻳﺲ و ﺷﻨﺎﺳﺎﻳﻲ ژنﻫﺎی ﻣﺮﺑﻮط ﺑﻪ اﻳﻦ ﺻﻔﺎت ﻣﻲﺗﻮان ﭘﺪﻳﺪه ﻫﺘﺮوزﻳﺲ را ﺑﺎ دﻳﺪﮔﺎه ﻣﻠﻜﻮﻟﻲ و ﺑﺎ ﺗﻤﺮﻛﺰ ﺑﺮ اﻛﺜﺮ ﺻﻔﺎت زراﻋﻲ و ﻓﻴﺰﻳﻮﻟﻮژﻳﻚ ﻣﺮﺗﺒﻂ ﺑﺎ اﻳﻦ ﭘﺪﻳﺪه، در ﭘﺮوژهﻫﺎی اﺻﻼح وارﻳﺘﻪﻫﺎی ﮔﻨﺪم ﺑﻪﻃﻮر ﻣﺆﺛﺮی وارد ﻛﺮد.

Comparision of wheat flag leaf to identify heterosis in resistance to septoria in wheat (Triticum aestivum)

Heterosis is controlled by genes/proteins that identification of these genes in molecular approach is important issue for plant breeding projects. This study was designed to investigate heterosis effect in resistance to septoria leaf blotch disease of wheat in 3 varieties, zagros as a sensitive, N8118 as a resistant parents and their hybrid respectively. The varieties were cultured in randomized complete block design in three repeats in Agricultural Research Station of Gorgan. Inoculation of plants was done at a time. To investigation of heterosis, the proteome analysis of flag leaf tissue was used; and protein extraction was done by using TCA/Aceton method. The results of proteome analysis before inoculation showed the differential expression of proteins between wheat hybrid and their parents in 5 spots. Also the presence of one protein spot in hybrid and the absence of that spot in parents after inoculation can be candidate protein of heterosis. The purpose of this research was improvement of molecular information about heterosis and using it in plant breeding projects in the most of agronomical and physiological traits related to this phenomon, specially resistant to septoria.
