چکیده
در ﺣﺎل ﺣﺎﺿﺮ ﮔﻮﻧﻪﻫﺎی ﻣﺨﺘﻠﻒ ﺳﭙﺘﻮرﻳﺎ ﺳﺎﻻﻧﻪ ﺣﺪود دو درﺻﺪ از ﻣﺤﺼﻮل ﮔﻨﺪم ﺟﻬﺎن را از ﺑﻴﻦ ﻣﻲﺑﺮﻧﺪ. در ﻫﻤﻴﻦ راﺳﺘﺎ از روش )ﺗﻜﻨﻴﻚ( ﭘﺮوﺗﺌﻮﻣﻴﻜﺲ ﺑﻪ ﻣﻨﻈﻮر ﺷﻨﺎﺳﺎﻳﻲ ﭘﺮوﺗﺌﻴﻦﻫﺎی ﭘﺎﺳﺦ دﻫﻨﺪه ﺑﻪ ﺑﻴﻤﺎری ﺳﭙﺘﻮرﻳﻮز ﺑﺮﮔﻲ، در ﮔﻨﺪم رﻗﻢ زاﮔﺮس اﺳﺘﻔﺎده ﮔﺮدﻳﺪ. ﺑﺪﻳﻦ ﻣﻨﻈﻮر ﺑﺬور رﻗﻢ زاﮔﺮس در اﻳﺴﺘﮕﺎه ﺗﺤﻘﻴﻘﺎت ﻛﺸﺎورزی ﮔﺮﮔﺎن در ﺳﻪ ﺗﻜﺮار و در ﻗﺎﻟﺐ ﻃﺮح آزﻣﺎﻳﺸﻲ ﺑﻠﻮک ﻛﺎﻣﻞ ﺗﺼﺎدﻓﻲ ﻛﺸﺖ ﮔﺮدﻳﺪ. ﻧﻤﻮﻧﻪ ﮔﻴﺮی از ﺑﺮگ ﭘﺮﭼﻢ ﻗﺒﻞ از آﻟﻮدﮔﻲ و ﺑﻌﺪ از آﻟﻮدﮔﻲ ﻣﺼﻨﻮﻋﻲ ﺑﻮﺗﻪﻫﺎ )ﺷﺎﺧﺺ ﻣﻘﺎوﻣﺖ ﺑﻪ ﻗﺎرچ( اﻧﺠﺎم ﮔﺮﻓﺖ. اﺳﺘﺨﺮاج ﭘﺮوﺗﺌﻴﻦ ﺑﻪ روش TCA/Aceton ﺗﻐﻴﻴﺮ ﻳﺎﻓﺘﻪ اﻧﺠﺎم ﮔﺮﻓﺖ و ﭘﺮوﺗﺌﻴﻦﻫﺎی اﺳﺘﺨﺮاج ﺷﺪه در دو ﺑﻌﺪ ﺑﻮﺳﻴﻠﻪ ژلﻫﺎی IPG
ﺟﺪاﺳﺎزی ﺷﺪﻧﺪ. ﻧﺘﺎﻳﺞ ﺣﺎﺻﻞ از ارزﻳﺎﺑﻲ ژلﻫﺎ ﻧﺸﺎن داد از ﻣﺠﻤﻮع 249 ﻧﻘﻄﻪ ﭘﺮوﺗﺌﻴﻨﻲ ﺗﻜﺮار ﭘﺬﻳﺮ ﺣﺪاﻗﻞ ﻧﻪ ﻧﻘﻄﻪ ﭘﺮوﺗﺌﻴﻨﻲ در ﻣﻘﺎوﻣﺖ ﺑﻪ ﺑﻴﻤﺎری ﺗﻐﻴﻴﺮات ﻣﻌﻨﻲ داری ﻧﺸﺎن دادﻧﺪ ﻛﻪ ﺑﻴﺎن ﺣﺪود 77 درﺻﺪ ﭘﺮوﺗﺌﻴﻦﻫﺎ اﻓﺰاﻳﺶ و 22 درﺻﺪ ﻛﺎﻫﺶ ﻳﺎﻓﺘﻪ ﺑﻮد. اﻳﻦ ﻧﺘﺎﻳﺞ ﻧﺸﺎن داد ﻛﻪ ﺑﺮرﺳﻲ اﻟﮕﻮی ﭘﺮوﺗﺌﻮم در ﻣﺮﺣﻠﻪ ﻗﺒﻞ و ﺑﻌﺪ از اﺳﭙﻮرﭘﺎﺷﻲ ﺣﺎﻛﻲ از اﻓﺰاﻳﺶ ﺗﻌﺪاد ﻟﻜﻪﻫﺎی ﭘﺮوﺗﺌﻴﻦ ﭘﺲ از اﺳﭙﻮرﭘﺎﺷﻲ ﻣﻲ ﺑﺎﺷﺪ. ﺑﻄﻮر ﻛﻠﻲ ﭼﻨﻴﻦ ﻧﺘﻴﺠﻪ ﮔﻴﺮی ﻣﻲﺷﻮد ﻛﻪ ﮔﻴﺎه در ﺑﺮاﺑﺮ ﻗﺎرچ ﺳﭙﺘﻮرﻳﺎ ﺑﺎ ﺑﻴﺎن ﭘﺮوﺗﺌﻴﻦﻫﺎی ﺟﺪﻳﺪ و اﻓﺰاﻳﺶ ﺑﻴﺎن ﭘﺮوﺗﺌﻴﻦﻫﺎی در ﺣﺎل ﺑﻴﺎن ﺑﻪ اﻳﻦ ﺑﻴﻤﺎری ﭘﺎﺳﺦ ﻣﻲدﻫﺪ.
ﺟﺪاﺳﺎزی ﺷﺪﻧﺪ. ﻧﺘﺎﻳﺞ ﺣﺎﺻﻞ از ارزﻳﺎﺑﻲ ژلﻫﺎ ﻧﺸﺎن داد از ﻣﺠﻤﻮع 249 ﻧﻘﻄﻪ ﭘﺮوﺗﺌﻴﻨﻲ ﺗﻜﺮار ﭘﺬﻳﺮ ﺣﺪاﻗﻞ ﻧﻪ ﻧﻘﻄﻪ ﭘﺮوﺗﺌﻴﻨﻲ در ﻣﻘﺎوﻣﺖ ﺑﻪ ﺑﻴﻤﺎری ﺗﻐﻴﻴﺮات ﻣﻌﻨﻲ داری ﻧﺸﺎن دادﻧﺪ ﻛﻪ ﺑﻴﺎن ﺣﺪود 77 درﺻﺪ ﭘﺮوﺗﺌﻴﻦﻫﺎ اﻓﺰاﻳﺶ و 22 درﺻﺪ ﻛﺎﻫﺶ ﻳﺎﻓﺘﻪ ﺑﻮد. اﻳﻦ ﻧﺘﺎﻳﺞ ﻧﺸﺎن داد ﻛﻪ ﺑﺮرﺳﻲ اﻟﮕﻮی ﭘﺮوﺗﺌﻮم در ﻣﺮﺣﻠﻪ ﻗﺒﻞ و ﺑﻌﺪ از اﺳﭙﻮرﭘﺎﺷﻲ ﺣﺎﻛﻲ از اﻓﺰاﻳﺶ ﺗﻌﺪاد ﻟﻜﻪﻫﺎی ﭘﺮوﺗﺌﻴﻦ ﭘﺲ از اﺳﭙﻮرﭘﺎﺷﻲ ﻣﻲ ﺑﺎﺷﺪ. ﺑﻄﻮر ﻛﻠﻲ ﭼﻨﻴﻦ ﻧﺘﻴﺠﻪ ﮔﻴﺮی ﻣﻲﺷﻮد ﻛﻪ ﮔﻴﺎه در ﺑﺮاﺑﺮ ﻗﺎرچ ﺳﭙﺘﻮرﻳﺎ ﺑﺎ ﺑﻴﺎن ﭘﺮوﺗﺌﻴﻦﻫﺎی ﺟﺪﻳﺪ و اﻓﺰاﻳﺶ ﺑﻴﺎن ﭘﺮوﺗﺌﻴﻦﻫﺎی در ﺣﺎل ﺑﻴﺎن ﺑﻪ اﻳﻦ ﺑﻴﻤﺎری ﭘﺎﺳﺦ ﻣﻲدﻫﺪ.

Study on expression of proteins involved in resistance to wheat ( Triticum aestivum ) flag leaf

Nowadays, different species of Septoria destroy about two percent of wheat production all over the world annually. Accordingly, proteomics method for identifying responding proteins to septoria leaf spotch disease in zagros variety of wheat was used. Hence, seeds of Zagros variety were cultured in complete randomized design model in three replicates in Agricultural Research Station of Gorgan. The sampling was done before inoculation of flag leaf tissue, the later sampling was done after appearance of the disease on the flag leaf. Protein extraction was done via TCA/Aceton method, and extracted proteins was separated in two dimension by IPG gels. Results, outcome from gel analysis of 249 repeatable protein spot showed that at least nine protein spot have significant difference in resistance to the disease, in which 77 percent of protein expression was increased and 22 percent decreased. These results showed that investigation of proteom pattern before and after of inoculation is indicative of increase in protein spot after inoculation. Generally, it is concluded that this variety respond to this disease by expressing new proteins and increasing proteins being expressed against to septoria.
