چکیده
ﺑﻪ ﻣﻨﻈﻮر ارزﻳﺎﺑﻲ اﺛﺮ ﻣﺘﻴﻞ ﺟﺎﺳﻤﻮﻧﺎت ﺑﺮ ﻓﻌﺎﻟﻴﺖ آﻧﺰﻳﻢﻫﺎی ﭘﻠﻲ ﻓﻨﻞ اﻛﺴﻴﺪاز و ﻓﻨﻴﻞ آﻻﻧﻴﻦ آﻣﻮﻧﻴﺎﻟﻴﺎز )PAL( در رﻗﻢ ﻛﺸﻜﻨﻲ ﻟﻮﻟﻮی رﻳﺤﺎن، آزﻣﺎﻳﺸﻲ در ﻗﺎﻟﺐ ﻃﺮح ﻛﺎﻣﻼ ﺗﺼﺎدﻓﻲ ﺑﺎ ﺳﻪ ﺗﻜﺮار در ﮔﻠﺨﺎﻧﻪ اﻧﺠﺎم ﺷﺪ. ﻣﺤﻠﻮل ﭘﺎﺷﻲ ﻣﺘﻴﻞ ﺟﺎﺳﻤﻮﻧﺎت در ﻣﺮﺣﻠﻪی ﭘﻴﺶ ﮔﻠﺪﻫﻲ )50 ﺑﺮﮔﻲ( ﺑﺮ روی ﺑﺮگﻫﺎی ﮔﻴﺎﻫﺎن ﺳﺎﻟﻢ اﻧﺠﺎم ﮔﺮدﻳﺪ. ﻏﻠﻈﺖ ﻣﺘﻴﻞ ﺟﺎﺳﻤﻮﻧﺎت در ﻣﻘﺎدﻳﺮ 0، 0/1 و 0/5 ﻣﻴﻠﻲ ﻣﻮﻻر ﺑﻪ ﻋﻨﻮان ﺗﻴﻤﺎر در ﻧﻈﺮ ﮔﺮﻓﺘﻪ ﺷﺪ. ﻣﻴﺰان ﻓﻌﺎﻟﻴﺖ آﻧﺰﻳﻢﻫﺎ 0، 24، 48 و 72 ﺳﺎﻋﺖ ﺑﻌﺪ از ﻣﺤﻠﻮل ﭘﺎﺷﻲ ﻣﺘﻴﻞ ﺟﺎﺳﻤﻮﻧﺎت اﻧﺪازهﮔﻴﺮی ﺷﺪ. ﺗﺠﺰﻳﻪ آﻣﺎری دادهﻫﺎ ﺑﺎ در ﻧﻈﺮ ﮔﺮﻓﺘﻦ زﻣﺎن ﺑﻪ ﻋﻨﻮان ﻋﺎﻣﻞ ﻓﺮﻋﻲ ﺑﺼﻮرت ﻃﺮح ﻛﺮتﻫﺎی ﺧﺮد ﺷﺪه اﻧﺠﺎم ﮔﺮﻓﺖ. ﻧﺘﺎﻳﺞ ﻧﺸﺎن داد ﻛﻪ ﺑﻴﺸﺘﺮﻳﻦ ﻓﻌﺎﻟﻴﺖ آﻧﺰﻳﻢ PAL در ﺗﻴﻤﺎر 0/5 ﻣﻴﻠﻲ ﻣﻮﻻر ﻣﺘﻴﻞ ﺟﺎﺳﻤﻮﻧﺎت ﻣﻲﺑﺎﺷﺪ. ﺑﻴﺸﺘﺮﻳﻦ ﻓﻌﺎﻟﻴﺖ اﻳﻦ آﻧﺰﻳﻢ در زﻣﺎن 24، 48 و ﻳﺎ 72 ﺳﺎﻋﺖ ﭘﺲ از ﻣﺤﻠﻮل ﭘﺎﺷﻲ ﻣﺸﺎﻫﺪه ﺷﺪ. ﺑﻴﺸﺘﺮﻳﻦ ﻓﻌﺎﻟﻴﺖ آﻧﺰﻳﻢ ﭘﻠﻲ ﻓﻨﻞ اﻛﺴﻴﺪاز در ﻏﻠﻈﺖ 0/1 ﻣﻴﻠﻲ ﻣﻮﻻر ﻣﺘﻴﻞ ﺟﺎﺳﻤﻮﻧﺎت و در زﻣﺎنﻫﺎی 24، 48 و 72 ﺳﺎﻋﺖ ﭘﺲ از ﻣﺤﻠﻮل ﭘﺎﺷﻲ ﻣﺸﺎﻫﺪه ﺷﺪ و ﺑﺎ اﻓﺰاﻳﺶ ﻏﻠﻈﺖ ﻣﺘﻴﻞ ﺟﺎﺳﻤﻮﻧﺎت ﻣﻴﺰان ﻓﻌﺎﻟﻴﺖ اﻳﻦ آﻧﺰﻳﻢ ﻛﺎﻫﺶ ﻳﺎﻓﺖ. ﻧﺘﺎﻳﺞ اﻳﻦ ﺗﺤﻘﻴﻖ ﻧﺸﺎن داد ﻛﻪ ﻣﻲﺗﻮان از ﻣﺤﻠﻮل ﭘﺎﺷﻲ ﻣﺘﻴﻞ ﺟﺎﺳﻤﻮﻧﺎت در اﻟﻘﺎی ﻓﻌﺎﻟﻴﺖ اﻳﻦ آﻧﺰﻳﻢﻫﺎ اﺳﺘﻔﺎده ﻧﻤﻮد.

Abstract
To evaluate the effect of methyl jasmonate on the activity of polyphenol oxidase and phenil alanin
amonialyase enzymes in basil (Ocimum basilicum L.) keshkeni luvelou cultivar, a completely randomized
design was conducted with three replications in greenhouse. Methyl jasmonate with different concentrations
of 0, 0.1 and 0.5 mM sprayed on the leaves in pre-flowering stage. The leaves of the sprayed plants were
sampled in 0, 24, 48, and 72 h after spraying. Different times after spraying was considered as sub-plots and
data was analyzed as split plot experiment. Results showed that the maximum activity of phenil alanin
amonialyase is in 0.5 mM concentration of methyl jasmonate. Also, the maximum activity of phenil alanin
amonialyase achieved 24, 48 or 72 h after spraying. The maximum activity of polyphenol oxidase was in 24,
48 and 72 h after spraying at 0.1 mM concentration of methyl jasmonate and increasing in methyl jasmonate
concentration declined the activity of the enzyme. In conclusion, it is concluded that use of methyl jasmonate
as an elicitor might induce the activity of these enzymes
