چکیده
ﺷﻨﺎﺳﺎﻳﻲ و ﻃﺒﻘﻪ ﺑﻨﺪی ﺑﻮﺗﻪ ﻫﺎی اﺳﺘﺒﺮق در ارﺗﻔﺎﻋﺎت ﻣﺨﺘﻠﻒ ﺑﺎ اﺳﺘﻔﺎده از ﻧﺸﺎﻧﮕﺮﻫﺎی ISSR ﺑﺮرﺳﻲ ﮔﺮدﻳﺪ. ﺑﻪ اﻳﻦ ﻣﻨﻈﻮر ﭘﺲ از اﺳﺘﺨﺮاج DNA، ﺑﻬﻴﻨﻪ ﺳﺎزی ﺷﺮاﻳﻂ و ﻏﻠﻈﺖﻫﺎی ﻣﺨﺘﻠﻒ ﻣﻮاد ﻣﻮرد اﺳﺘﻔﺎده در PCR، 9 آﻏﺎزﮔﺮ ﺑﻪ ﺻﻮرت اﻧﻔﺮادی و ﺗﺮﻛﻴﺒﻲ ﺑﻪ ﻛﺎر ﮔﺮﻓﺘﻪ ﺷﺪﻧﺪ. در ﻣﺠﻤﻮع 259 ﻗﻄﻌﻪ ﺗﻜﺜﻴﺮ ﺷﺪ ﻛﻪ 106 ﻗﻄﻌﻪ ازآﻧﻬﺎ ﭼﻨﺪ ﺷﻜﻞ ﺑﻮدﻧﺪ. ﻣﺤﺎﺳﺒﺎت آﻣﺎری ﺑﺎ اﺳﺘﻔﺎده از روش ﺟﺎﻛﺎرد و ﻧﺮماﻓﺰار NTSYS Ver.2.02 اﻧﺠﺎم ﮔﺮﻓﺖ. ﻣﻴﺰان PIC ﺑﺮای آﻏﺎزﮔﺮﻫﺎی ﻣﺨﺘﻠﻒ از 0/37 ﺑﺮای آﻏﺎزﮔﺮ ISSR2 ﺗﺎ 0/46 ﺑﺮای آﻏﺎزﮔﺮﻫﺎی ISSR5 و ISSR9 ﻣﺘﻐﻴﺮ ﺑﻮد.14 ﻧﻤﻮﻧﻪ در ﺳﻪ ﮔﺮوه ﺟﺎی ﮔﺮﻓﺘﻨﺪ ﻛﻪ 2 ﮔﺮوه ﻫﺮﻛﺪام 6 ﻧﻤﻮﻧﻪ ، و ﻳﻚ ﮔﺮوه 3 ﻧﻤﻮﻧﻪ را ﺟﺎی دادﻧﺪ. در ﻣﺠﻤﻮع ﺗﻜﻨﻴﻚ ISSR ﺑﺮای ﻃﺒﻘﻪ ﺑﻨﺪی ﮔﻴﺎﻫﺎن اﺳﺘﺒﺮق ﻣﻔﻴﺪ اﺳﺖ و ﺷﺎﻳﺪ اﺳﺘﻔﺎده از آﻏﺎزﮔﺮﻫﺎی زﻳﺎدﺗﺮ ﻛﻤﻚ ﺑﻴﺸﺘﺮی ﺑﺮای ﺗﻔﻜﻴﻚ و ﮔﺮوهﺑﻨﺪی ارﻗﺎم ﻧﻤﺎﻳﺪ. ﺗﻨﻮع ژﻧﺘﻴﻜﻲ ﻗﺎﺑﻞ ﺗﻮﺟﻬﻲ درون اﺳﺘﺒﺮق ﻫﺎی ﺑﻮﻣﻲ اﺳﺘﺎن ﻛﺮﻣﺎن وﺟﻮد دارد و ﻧﻤﻮﻧﻪ ﻫﺎی اﺳﺘﺒﺮق از ﻧﻈﺮ ﺳﻄﺢ ﺗﻨﻮع ژﻧﺘﻴﻜﻲ ﺑﺎ ﻳﻜﺪﻳﮕﺮ ﻣﺘﻔﺎوت ﻫﺴﺘﻨﺪ. ﻋﻮاﻣﻞ ﻣﺤﻴﻄﻲ و ﺗﻜﺜﻴﺮ ﺟﻨﺴﻲ اﺣﺘﻤﺎﻻ دﻟﻴﻞ اﺻﻠﻲ اﻳﺠﺎد ﻓﺎﺻﻠﻪ ژﻧﺘﻴﻜﻲ ﺑﻴﻦ ﻧﻤﻮﻧﻪ ﻫﺎ ﻣﻲ ﺑﺎﺷﻨﺪ.

Using ISSR Markers in Genetic Diversity Milkweed

Abstract
Identification and classification of milkweed plants in different heights using ISSR markers were studied. For this purpose, next to DNA extraction, optimization of Conditions and Different concentrations of materials used in the PCR, 9 primers were used individually and in combination. in total, 259 Pieces were amplified that, 106 of them were polymorphic fragments. Statistical analysis was performed using jakard method and NTSYS Ver.2.02 software. PIC levels for different markers , were variable from 0.37 for ISSR2 marker to 0/46 for ISSR5 and ISSR9 markers. 14 samples were classified into three groups that 2 groups have 6 samples and 1 group has 3 samles. In total, ISSR technique is useful for classifying milkweed plants and perhaps more helpful for identification and grouping of samples, using more markers. Considerable genetic variation exists within kerman native milkweeds and Milkweed examples are different in levels of genetic diversity. Likely The main reason for the creation of genetic distance Between samples are Environmental factors and sexual reproduction.
