چکیده
ﻛﻠﭙﻮره(Teucrium poliumL) ﻳﻜﻲ از ﻣﻬﻤﺘﺮﻳﻦ ﮔﻴﺎﻫﺎن داروﻳﻲ اﺳﺖ ﻛﻪ ﺑﻴﺶ از ﻫﺰار ﺳﺎل اﺳﺖ ﻛﻪ در ﻃﺐ ﺳﻨﺘﻲ اﺳﺘﻔﺎده ﻣﻲﺷﻮد و دارای ﺧﺼﻮﺻﻴﺎت داروﻳﻲ ﻣﺘﻌﺪد ﻣﺎﻧﻨﺪ ﺿﺪ دﻳﺎﺑﺖ و ﺿﺪ ﺳﺮﻃﺎن اﺳﺖ. ﺗﻮﻟﻴﺪ رﻳﺸﻪﻫﺎی ﻣﻮﻳﻴﻦ ﻳﻚ ﺳﻴﺴﺘﻢ ارزﺷﻤﻨﺪ ﺟﻬﺖ ﻣﻄﺎﻟﻌﺎت ﺑﻴﻮﺳﻨﺘﺰ ﻣﺘﺎﺑﻮﻟﻴﺖﻫﺎی ﺛﺎﻧﻮﻳﻪ و ﺗﻮﻟﻴﺪ در ﻣﻘﻴﺎس وﺳﻴﻊ ﺗﺮﻛﻴﺒﺎت داروﻳﻲ اﺳﺖ. در اﻳﻦ ﺗﺤﻘﻴﻖ، رﻳﺰﻧﻤﻮﻧﻪﻫﺎی ﻣﺨﺘﻠﻒ ﺟﺪا ﺷﺪه از T. polium ﺑﺮای اﻟﻘﺎی رﻳﺸﻪ ﻣﻮﻳﻴﻦ ﺑﺎ اﺳﺘﻔﺎده از ﺗﺮارﻳﺨﺘﻲ ژﻧﺘﻴﻜﻲ ﺑﺎ دو ﺳﻮﻳﻪ (A13 و A7)ﺑﺎﻛﺘﺮی Agrobacterium rhizogenes اﺳﺘﻔﺎده ﺷﺪﻧﺪ. ﺗﺮارﻳﺨﺘﻲ رﻳﺸﻪﻫﺎ ﺑﺎ اﺳﺘﻔﺎده از واﻛﻨﺶ PCR ﺑﺮای ﺗﻜﺜﻴﺮاﺧﺘﺼﺎﺻﻲ ژن rolA-B ﺗﺄﻳﻴﺪ ﺷﺪ. ﺑﻴﺸﺘﺮﻳﻦ ﺗﻌﺪاد رﻳﺸﻪ اﻟﻘﺎﻳﻲ )7/58 رﻳﺸﻪ در ﻫﺮ رﻳﺰﻧﻤﻮﻧﻪ( زﻣﺎﻧﻲ ﺣﺎﺻﻞ ﺷﺪ ﻛﻪ رﻳﺰﻧﻤﻮﻧﻪﻫﺎی ﮔﻴﺎﻫﭽﻪ 32 روزه ﺑﻪ دﺳﺖ آﻣﺪه از ﮔﻴﺎﻫﭽﻪﻫﺎی درون ﺷﻴﺸﻪای، ﺑﺎ ﺳﻮﻳﻪ A7ﺗﺮارﻳﺨﺖ ﺷﺪﻧﺪ .

Optimizing the induction of hairy roots by Agrobacterium rhizogenes into Teucrium polium L.

Abstract
Teucrium polium L. is one of the most important herbs that have been used over thousands of years in traditional medicine and has multiple medicinal usages: anti-diabetic and anti-cancer, etc. Hairy roots production of a valuable system is for studying the biosynthesis of secondary metabolites and for large-scale production of pharmaceutical compounds. In this research, several explants isolated from T. polium were used to induce the hairy root, using genetic transformation with two-strains (A13 and A7) Agrobacterium rhizogenes. Root transformation was confirmed using PCR reactions to specifically amplify rolA-B gene. Maximum number of root inductions (7.58 roots per explant) were achieved just when the 32-day-old seedling explants derived from in vitro seedlings, transformed with A7 strain.
