چکیده
زﻋﻔﺮان از ﺟﻤﻠﻪ ﮔﻴﺎﻫﺎن ﺑﻮﻣﻲ اﻳﺮان ﺑﻮده ﻛﻪ دارای ﺧﻮاص داروﻳﻲ و ادوﻳﻪای ﻣﺘﻌﺪد ﻣﻲﺑﺎﺷﺪ و ﺑﻪ ﻟﺤـﺎظ اﻗ ﺘﺼـﺎدی در اﻳـﺮان و ﺟﻬـﺎن ﺣﺎﺋﺰ اﻫﻤﻴﺖ اﺳﺖ. ﻳﻜﻲ از ﻛﺎرﺑﺮدﻫﺎی ﻣﻬﻢ ﺗﻜﻨﻴﻚﻫﺎی ﻣﻮﻟﻜﻮﻟﻲ ﺑﺮرﺳﻲ و ﺑﺮآورد ﺳﻄﺢ ﺗﻨﻮع ژﻧﺘﻴﻜﻲ در ژرمﭘﻼﺳﻢﻫـﺎ و ﺟﻤﻌﻴـﺖ ﻫـﺎ،ﺑﺮای اﺳﺘﻔﺎده ﻣﻄﻠﻮب از آنﻫﺎ در ﺑﻪﻧﮋادی و ﻣﺪﻳﺮﻳﺖ ﺣﻔﻆ ذﺧﺎﻳﺮ ژﻧﺘﻴﻜﻲ ﮔﻴﺎﻫﺎن ﻣﻲﺑﺎﺷﺪ. ﺑﻪ اﻳﻦ ﻣﻨﻈﻮر ﺗﻨﻮع و ارﺗﺒﺎط ژﻧﺘﻴﻜﻲ 25 ﻧﻤﻮﻧﻪ از زﻋﻔﺮانﻫﺎی زراﻋﻲ و وﺣﺸﻲ ﺷﺎﻣﻞ ﮔﻮﻧﻪﻫﺎی C. haussknechii ،C. cansellatuse ،C. speciosus ،C. sativus و C. michelsonii ﺑﺎ اﺳﺘﻔﺎده از ﭼﻨﺪ ﻧﺸﺎﻧﮕﺮ RAPD ﻣﻮرد ﺑﺮرﺳﻲ ﻗﺮار ﮔﺮﻓـﺖ. ﻳـﺎزده آﻏـﺎزﮔﺮ RAPD 90 ﻧـﻮار ﻗﺎﺑـﻞ اﻣﺘﻴـ ﺎزدﻫﻲ ﺗﻮﻟﻴـﺪ ﻛﺮدﻧـﺪ. ﻣﻴـﺎﻧﮕﻴﻦ ﭼﻨﺪﺷﻜﻠﻲ ﺑﻪ دﺳﺖ آﻣﺪه ﺑﺮای ﻧﺸﺎﻧﮕﺮ RAPD ﻣﻌﺎدل 77/95 درﺻﺪ ﻣﺤﺎﺳﺒﻪ ﺷﺪ. ﻧﺘﺎﻳﺞ ﺣﺎﺻﻞ از ﺗﺠﺰﻳﻪ وارﻳﺎﻧﺲ ﻣﻮﻟﻜﻮﻟﻲ ﻧﺸﺎن داد ﻛـﻪ ﺗﻨﻮع ژﻧﺘﻴﻜﻲ ﺑﻴﻦ ﺟﻤﻌﻴﺖﻫﺎ و درون ﺟﻤﻌﻴﺖﻫﺎ ﻣﻌﻨﻲدار ﺑﻮد. ﺑﺮ اﺳﺎس ﻧﺘﺎﻳﺞ ﺣﺎﺻﻞ از ﺗﺠﺰﻳﻪ وارﻳﺎﻧﺲ ﻣﻮﻟﻜﻮﻟﻲ اﻳﻦ ﺗﺠﺰﻳﻪ ﺳﻬﻢ ﺗﻨـﻮع ژﻧﺘﻴﻜﻲ ﺑﻴﻦ ﺟﻤﻌﻴﺖﻫﺎ و درون ﺟﻤﻌﻴﺖﻫﺎ ﺑﻪ ﺗﺮﺗﻴﺐ ﺑﺮاﺑﺮ ﺑﺎ 36 و 64 درﺻﺪ ﺑﺮآورد ﮔﺮدﻳﺪ. ﺗﻮزﻳﻊ ﻣﻘﺎدﻳﺮ PIC ﺑﻴﻦ 0/119 ﺗﺎ 0/313 ﺑـﺎ ﻣﻴﺎﻧﮕﻴﻦ 0/257 ﻣﺘﻐﻴﺮ ﺑﻮد. ﻧﺘﺎﻳﺞ ﺣﺎﺻﻞ ﻧﺸﺎن داد ﻛﻪ در ﺑﻴﻦ و درون ﮔﻮﻧﻪﻫﺎی زﻋﻔﺮان ﺗﻨﻮع زﻳﺎدی وﺟـﻮد دارد ﻛـﻪ ﻣـ ﻲﺗـﻮان از آن در ﻧﺎﻣﻪﻫﺎی ﺑﻪ ﻧﮋادی اﻳﻦ ﮔﻴﺎه ﺑﺎ ارزش اﺳﺘﻔﺎده ﻛﺮد.

Evaluation of genetic diversity and phylogenetic relationships in some species of saffron using RAPD markers

Abstract
Saffron (Crocus sativa L.) is a native plant of Iran with various medical and food properties. It also has an important role in terms of economical issues in Iran. One of the major applications of molecular techniques to investigate and estimate the level of genetic diversity in germplasm and populations, it is ideal for use in breeding and management of plant genetic resources is maintained. The diversity and genetic relationship of 25 samples of cultivated and wild species of crocus C. sativus, C. speciosus, C. cansellatuse, C. haussknechii and C. michelsonii were studied using several RAPD markers. Eleven RAPD primers produced 90 bands it scored. Average polymorphism of RAPD markers obtained 77. 95 %. Results of analysis of molecular variance showed that genetic variation between populations and within populations were significant. AMOVA analysis showed that the variation between the species, between different populations, and within populations accounted for 36, and 64% of the total variation, respectively. Mean polymorphic information content (PIC) of the markers was 0.257 and ranged between 0.119 and 0.313. The results showed that there is a lot of variation between and within species crocus that can be used to value the plant breeding programs.
