چکیده
ﺑﺬرﻫﺎی ﻛﺪوی ﺗﺨﻢ ﻛﺎﻏﺬی ﻣﻨﺒﻊ ﺳﺮﺷﺎری از ﭘﺮوﺗﺌﻴﻦ و روﻏﻦ اﺳﺖ و ﻣﻮاد ﻣﻮﺛﺮه ارزﺷﻤﻨﺪی از ﺟﻤﻠﻪ اﺳﻴﺪﻫﺎی ﭼﺮب، ﻓﻴﺘﻮﺳـﺘﺮول و وﻳﺘﺎﻣﻴﻦ E )ﺑﻪوﻳﮋه ﮔﺎﻣﺎ ﺗﻮﻛﻮﻓﺮول( دارد. ﺑﻪ ﻣﻨﻈﻮر ارزﻳﺎﺑﻲ اﺛﺮ اﻧﻮاع و ﻣـﺪت ﭘـ ﻴﺶ ﺗﻴﻤـﺎر ﺑـﺮ روی ﺗﻘﻮﻳـﺖ ﺑـﺬر و ﺑﺮﺧـﻲ از ﺻـﻔﺎت ﺑﻴﻮﺷﻴﻤﻴﺎﻳﻲ ﺟﻮاﻧﻪزﻧﻲ، آزﻣﺎﻳﺸﻲ ﺑﻪ ﺻﻮرت ﻓﺎﻛﺘﻮرﻳﻞ ﺑﺮ ﭘﺎﻳﻪ ﻃﺮح ﻛﺎﻣﻼ ﺗﺼﺎدﻓﻲ ﺑﺎ ﺳﻪ ﺗﻜﺮار در داﻧﺸﮕﺎه ﻣﺤﻘﻖ اردﺑﻴﻠـﻲ اﺟـﺮا ﮔﺮدﻳـﺪ ﺗﻴﻤﺎرﻫﺎی آزﻣﺎﻳﺶ ﺷﺎﻣﻞ ﭘﻴﺶ ﺗﻴﻤﺎر ﺑﺎ آب و ﻣﺤﻠﻮلﻫﺎی ﺳﻪ درﺻﺪ NaCl، 20 ﻣﻴﻠﻲﮔﺮم ﺑـﺮ ﻟﻴﺘـﺮ اﺳـﻴﺪ ﺟ ﻴﺒﺮﻟﻴـﻚ وppm 20 اﺳـﻴﺪ آﺳﻜﻮرﺑﻴﻚ ﺑﻪ ﻣﺪتﻫﺎی 8، 16و 24 ﺳﺎﻋﺖ ﺑﻮد. ﻳﻚ ﺗﻮده ﺑﺬری ﺑﻪ ﻋﻨﻮان ﺷﺎﻫﺪ ﺑﺮای ﻣﻘﺎﻳﺴﻪ روشﻫﺎی ﻣﺨﺘﻠـﻒ ﭘـﻴﺶ ﺗﻴﻤـﺎر در ﻧﻈـﺮ ﮔﺮﻓﺘﻪ ﺷﺪ. ﻧﺘﺎﻳﺞ آزﻣﺎﻳﺶ ﻧﺸﺎن داد ﻛﻪ در ﺑﺬرﻫﺎی ﭘﺮاﻳﻢ ﺷﺪه ﺑﺎ آب، اﺳﻴﺪ ﺟﻴﺒﺮﻟﻴﻚ و اﺳﻴﺪ آﺳﻜﻮرﺑﻴﻚ، درﺻﺪ و ﺳﺮﻋﺖ ﺟﻮاﻧﻪزﻧـﻲ ﺑـﺎ اﻓﺰاﻳﺶ ﻣﺪت زﻣﺎن ﭘﺮاﻳﻤﻴﻨﮓ اﻓﺰاﻳﺶ ﻧﺸﺎن داد ﻛﻪ اﻳﻦ ﻣﻴﺰان اﻓﺰﻳﺶ در ﺗﻴﻤﺎر اﺳﻴﺪ آﺳﻜﻮرﺑﻴﻚ ﻣﺤﺴﻮسﺗﺮ ﺑﻮد. آﻧﺰﻳﻢﻫـﺎی ﺳﻮﭘﺮاﻛﺴـﻴﺪ دﻳﺴﻤﻴﻮﺗﺎز، ﻛﺎﺗﺎﻻز و ﭘﺮاﻛﺴﻴﺪاز در ﺗﻴﻤﺎرﻫﺎی اﺳﻴﺪ ﺟﻴﺒﺮﻟﻴﻚ و ﻛﻠﺮﻳﺪ ﺳﺪﻳﻢ ﺑﻪ ﺗﺮﺗﻴﺐ ﺑﻴﺸﺘﺮﻳﻦ و ﻛﻤﺘـﺮﻳﻦ ﻣﻘـﺪار ﻓﻌﺎﻟﻴـﺖ را ﺑـﻪ ﺧـﻮد اﺧﺘﺼﺎص دادﻧﺪ. ﻓﻌﺎﻟﻴﺖ اﻳﻦ آﻧﺰﻳﻢﻫﺎ ﺑﻪ ﻏﻴﺮ از ﻫﺎﻟﻮ ﭘﺮاﻳﻤﻴﻨﮓ ﺑﺎ اﻓﺰاﻳﺶ ﻣﺪت زﻣﺎن ﭘﺮاﻳﻤﻴﻨﮓ روﻧﺪ ﺻﻌﻮدی را ﻧﺸﺎن داد، ﺑﻪ ﻃﻮری ﻛـﻪ ﺑﺬرﻫﺎی ﭘﻴﺶ ﺗﻴﻤﺎر ﺷﺪه ﺑﺎ اﺳﻴﺪ ﺟﻴﺒﺮﻟﻴﻚ ﺑﻪ ﻣﺪت 24 ﺳﺎﻋﺖ ﻣﻨﺎﺳﺐﺗﺮﻳﻦ ﺗﻴﻤﺎر از ﻧﻈﺮ ﻣﻴﺰان ﻓﻌﺎﻟﻴﺖ اﻳﻦ آﻧﺰﻳﻢ ﺑﻮدﻧﺪ.

Pumpkin seed oil is a good source of protein and valuable active ingredients such as fatty acids, phytosterols and vitamin E (especially gamma tocopherol). In order to evaluate the effects of priming materials and duration of priming on the seed physiological enhancement and biochemical traits of germination in pumpkin seed a factorial experiment conducted based on completely randomized desigh with three replications at the University of MohagheghArdabili. Treatments were priming with water, NaCl (3%), gibberellic acid (20 mg L-1) and ascorbic acid (20 ppm)to periods of 8, 16 and 24 hours. Seed mass as a control for comparison of different pretreatment methods were considered. Percentage and germination rate in the seeds primed with water, gibberellic acid and ascorbic acid increased with increasing duration of priming showed that the rate of increase was even higher in ascorbic acid treatment. Superoxide dismutase, catalase and peroxidase in sodium chloride and gibberellic acid treatments, had the highest and lowest activity respectively. The activity of these enzymes had upward trend except in halo-priming, and hormone-priming with gibberellin for 24 h was the best treatment considering enzyme activity.
