چکیده
ﺑﻪ ﻣﻨﻈﻮر ﺑﺮرﺳﻲ ﭘﺎﺳﺦ ﺑﻪ ﺗﻨﺶ اﻛﺴﻴﺪاﺗﻴﻮ ﻧﺎﺷﻲ از ﻛﺎدﻣﻴﻮم در ارﻗﺎم ﻣﻘﺎوم و ﺣﺴﺎس ﮔﻠﺮﻧﮓ )Carthamus tinctorius L.( آزﻣﺎﻳﺸﻲ ﻓﺎﻛﺘﻮرﻳﻞ و در ﻗﺎﻟﺐ ﻃﺮح ﺑﻠﻮکﻫﺎی ﻛﺎﻣﻞ ﺗﺼﺎدﻓﻲ ﺑﺎ ﭼﻬﺎر ﺗﻜﺮار در داﻧﺸﻜﺪه زﻳﺴﺖ ﺷﻨﺎﺳﻲ داﻧﺸﮕﺎه اﺳﺘﻜﻬﻠﻢ ﺳﻮﺋﺪ و در اﺗﺎﻗﻚ رﺷﺪ در ﻣﺤﻴﻂ ﻛﺸﺖ ﻫﻴﺪروﭘﻮﻧﻴﻚ ﺑﺎ ﺷﺮاﻳﻂ ﻛﺎﻣﻼ ﻛﻨﺘﺮل ﺷﺪه ﻣﻮرد ﺑﺮرﺳﻲ ﻗﺮار ﮔﺮﻓﺖ. ﺳﻪ ﺳﻄﺢ آﻟﻮدﮔﻲ ﺑﻪ ﻛﺎدﻣﻴﻮم 0، 1، 20 ﻣﻴﻜﺮوﻣﻮﻻر( ﺑﺮ ﺳﻪ ژﻧﻮﺗﻴﭗ ﮔﻠﺮﻧﮓ اﻋﻤﺎل ﮔﺮدﻳﺪ. ﭘﺎراﻣﺘﺮﻫﺎی ﺷﺎﺧﺺ ﻣﻘﺎوﻣﺖ ﺑﻪ ﻛﺎدﻣﻴﻮم، ﭘﺮاﻛﺴﻴﺪاﺳﻴﻮن ﻟﻴﭙﻴﺪ ﺑﺮگ،ﻧﺴﺒﺖ ﻛﺎدﻣﻴﻮم ﺑﺎﻧﺪ ﺷﺪه ﺑﺎ دﻳﻮاره ﺳﻠﻮﻟﻲ رﻳﺸﻪ، ﻣﻴﺰان ﻓﻌﺎﻟﻴﺖ آﻧﺰﻳﻢ ﺳﻮﭘﺮاﻛﺴﻴﺪ دﺳﻤﻮﺗﺎز )SOD( و ﻣﻜﺎن ﺗﺸﻜﻴﻞ ﺳﻮﺑﺮﻳﻦ ﻻﻣﻼ ﭘﺲ از دو ﻫﻔﺘﻪ ﻗﺮار ﮔﺮﻓﺘﻦ در ﻣﺤﻴﻂ آﻟﻮده اﻧﺪازهﮔﻴﺮی ﺷﺪﻧﺪ. ﺑﺎ ﺗﻮﺟﻪ ﺑﻪ ﺷﺎﺧﺺ ﻣﻘﺎوﻣﺖ ﺑﻪ ﻛﺎدﻣﻴﻮم و ﭘﺮاﻛﺴﻴﺪاﺳﻴﻮن ﻟﻴﭙﻴﺪﻫﺎ ژﻧﻮﺗﻴﭗ AC-Sterling، اراک و Saffire ﺑﻪ ﺗﺮﺗﻴﺐ ﺑﻪ ﻋﻨﻮان ژﻧﻮﺗﻴﭙﻬﺎی ﻣﻘﺎوم، ﺣﺴﺎس و ﻧﻴﻤﻪ ﺣﺴﺎس ﺑﻪ ﻛﺎدﻣﻴﻮم ﺷﻨﺎﺧﺘﻪ ﺷﺪﻧﺪ. ﺑﻴﺸﺘﺮﻳﻦ و ﻛﻤﺘﺮﻳﻦ ﻏﻠﻈﺖ ﻛﺎدﻣﻴﻮم ﺑﺎﻧﺪ ﺷﺪه ﺑﺎ دﻳﻮاره ﺳﻠﻮﻟﻲ رﻳﺸﻪ در ﺳﻄﺢ 1 ﻣﻴﻜﺮوﻣﻮﻻر ﺑﻪ ﺗﺮﺗﻴﺐ ﻣﺮﺑﻮط ﺑﻪ AC-Sterling و اراک ﺑﻮد، درﺣﺎﻟﻴﻜﻪ در ﺳﻄﺢ 20 ﻣﻴﻜﺮوﻣﻮﻻر ژﻧﻮﺗﻴﭙﻬﺎ ﺑﻪ ﻟﺤﺎظ اﻳﻦ ﺻﻔﺖ ﺗﻔﺎوت ﻣﻌﻨﻲدار ﻧﺪاﺷﺘﻨﺪ. اوج ﻓﻌﺎﻟﻴﺖ آﻧﺰﻳﻢ SOD ﺑﺮگ ژﻧﻮﺗﻴﻬﺎی Saffire و اراک در ﺳﻄﺢ 1 و AC-Sterling در 20 ﻣﻴﻜﺮوﻣﻮﻻر ﻣﺸﺎﻫﺪه ﺷﺪ. ﻫﻤﭽﻨﻴﻦ ﻣﻜﺎن ﺗﺸﻜﻴﻞ ﺳﻮﺑﺮﻳﻦ ﻻﻣﻼ ﺑﻴﻦ ژﻧﻮﺗﻴﭙﻬﺎی ﻣﻮرد ﻣﻄﺎﻟﻌﻪ ﻣﺘﻔﺎوت ﻧﺒﻮد.

Evaluation of some physiological responses of tolerant and non tolerant safflower genotypes to Cd-inuced oxidative stress

Abstract
In order to investigate the physiological responses of tolerant and non-tolerant safflower genotypes to Cd-induced oxidative stress, a 4-replicate randomized complete block factorial experiment was conducted at the Stockholm University, Sweden. three levels of Cd (including 0, 1 and 20 µM CdCl2) and three genotypes (AC-Sterling, Arak and Saffire) were applied to the hydroponics culture condition. Cd tolerance index (CTI), lipid peroxidation, superoxide dismutase (SOD) activities and suberization associated with the casparian band in the root were measured in 14-day treated plants. According to the Both CTI and lipid peroxidation AC-Sterling and Arak were tolerant and non-tolerant genotypes respectively. Cell wall-bound Cd in safflower root did not indicate a significant role in genotypic differences at least under the high CdCl2 levels in the nutrient solution. Cell wall-bound Cd in safflower root, however, did appear to play a role under the low CdCl2 levels at least with the tolerant Ac-Sterling. In this study, however, no root suberzation difference was observed between the three genotypes. The SOD activity in response to Cd stress was genotype- and Cd-dependent. The non-tolerant and semi-tolerant genotypes did not indicate a high SOD activity under high Cd levels.
