ﺑﺮرﺳﻲ ﺗﻨﻮع ژﻧﺘﻴﻜﻲ ﺗﻮدهﻫﺎی اﺳﻔﻨﺎج ﺑﻮﻣﻲ اﻳﺮان(Spinacia oleracea L) ﺑﺎ اﺳﺘﻔﺎده از ﻧﺸﺎﻧﮕﺮ ﻣﻮﻟﻜﻮﻟﻲ رﻳﺰﻣﺎﻫﻮاره

چکیده
ﺗﻌﻴﻴﻦ ﻣﻴﺰان ﺗﻨﻮع ژﻧﺘﻴﻜﻲ در ذﺧﺎﻳﺮ ﺗﻮارﺛﻲ ﮔﻴﺎﻫﻲ اوﻟﻴﻦ ﻗﺪم در ﺑﺮﻧﺎﻣﻪﻫﺎی اﺻﻼح ﻧﺒﺎﺗﺎت اﺳﺖ. ﺑﻪ ﻣﻨﻈﻮر ﺗﻌﻴﻴﻦ ﺗﻨﻮع ژﻧﺘﻴﻜﻲ، 20 رﻗﻢ اﺳﻔﻨﺎج اﻳﺮاﻧﻲ در 5 ﺗﻜﺮار ﺑﺎ اﺳﺘﻔﺎده از ﻧﺸﺎﻧﮕﺮﻫﺎی رﻳﺰﻣﺎﻫﻮاره ﻣﻮرد ﻣﻄﺎﻟﻌﻪ ﻗﺮار ﮔﺮﻓﺘﻨﺪ. در اﻳﻦ ﻣﻄﺎﻟﻌﻪ 10 ﺟﻔﺖ آﻏﺎزﮔﺮ ﺑﻜﺎر ﮔﺮﻓﺘﻪ ﺷﺪ. 2 ﺟﻔﺖ از آﻏﺎزﮔﺮﻫﺎی اﺳﺘﻔﺎده ﺷﺪه ﺑﻪ ﻋﻠﺖ ﻋﺪم ﺗﻜﺜﻴﺮ ﺑﺎﻧﺪ از اداﻣﻪ آزﻣﺎﻳﺸﺎت ﺣﺬف ﺷﺪﻧﺪ. ﺳﻴﺴﺘﻢ ﻣﺎرﻛﺮ در ﻣﺠﻤﻮع ﺗﻮاﻧﺴﺖ 42 آﻟﻞ را ﺑﺎ اﻧﺪازهی ﺑﻴﻦ 280- 110 ﺟﻔﺖ ﺑﺎز ﺷﻨﺎﺳﺎﻳﻲ ﻛﻨﻨﺪ. آﻏﺎزﮔﺮ X80044 ،X17631 و U33330 ﺑﺎ 7 آﻟﻞ و آﻏﺎزﮔﺮ X66559 ﺑﺎ اﻳﺠﺎد 4 آﻟﻞ ﺑﻪ ﺗﺮﺗﻴﺐ ﺑﻴﺸﺘﺮﻳﻦ و ﻛﻤﺘﺮﻳﻦ ﺗﻌﺪاد آﻟﻞ را داﺷﺘﻨﺪ. ﻣﻴﺎﻧﮕﻴﻦ ﺗﻌﺪاد آﻟﻞﻫﺎی آﺷﻜﺎر ﺷﺪه ﺑﻪ ازای ﻫﺮ ﺟﺎﻳﮕﺎه 4/963 ﺑﺮآورد ﺷﺪ. ﺑﻴﺸﺘﺮﻳﻦ ﻓﺮاواﻧﻲ آﻟﻠﻲ ﻣﺮﺑﻮط ﺑﻪ آﻟﻞ 4 ﺟﺎﻳﮕﺎه X17031، ﺑﺎ ﻓﺮاواﻧﻲ 0/460 و. ﻛﻤﺘﺮﻳﻦ ﻓﺮاواﻧﻲ آﻟﻠﻲ ﻣﺸﺎﻫﺪه ﺷﺪه 0/005 ﻣﺮﺑﻮط ﺑﻪ آﻟﻞ 6 ﺟﺎﻳﮕﺎه X17162 ﻣﻲﺑﺎﺷﺪ. ﺑﻴﺸﺘﺮﻳﻦ و ﻛﻤﺘﺮﻳﻦ ﻣﻴﺰان اﻃﻼﻋﺎت ﭼﻨﺪﺷﻜﻠﻲ ﺑﺎ ﻣﻘﺪار 0/839 و 0/703 ﺑﻪ ﺗﺮﺗﻴﺐ ﻣﺮﺑﻮط ﺑﻪ آﻏﺎزﮔﺮﻫﺎی X80044 و Z66559 ﻣﺸﺎﻫﺪه ﮔﺮدﻳﺪ. ﻣﻴﺎﻧﮕﻴﻦ PIC ﺑﻪ دﺳﺖ آﻣﺪه ﺑﺮای ﺗﻤﺎم ﺟﺎﻳﮕﺎهﻫﺎ ﺑﺮاﺑﺮ ﺑﺎ 0/774 ﺑﻮد ﻛﻪ ﻫﺘﺮوزﻳﮕﻮﺳﻴﺘﻲ ﺑﺎﻻ را در اﻳﻦ ﺗﻮدهﻫﺎ ﻧﺸﺎن داد. ﮔﺮوهﺑﻨﺪی ﺑﺎ اﺳﺘﻔﺎده از ﺿﺮﻳﺐ ﺗﺸﺎﺑﻪ ﻧﻲ و اﻟﮕﻮرﻳﺘﻢ UPGMA، ﺗﻮدهﻫﺎ را در ﭼﻬﺎر ﮔﺮوه ﻗﺮار داد.

Abstract
Determine the extent of genetic variation in the inheritance of plant resources is the first step in breeding
programs. In order to determine the genetic diversity of 20 Iranian spinach cultivars using microsatellite
markers were studied in five replicates. In this study, 10 primer pairs were used. 2 pairs of primers were used
in the experiments were removed from the band due to non-proliferation. Marker systems have a total of 42
alleles with a size between 110-280 bp to detect. Primers X17631, X80044 and U33330 7 allele and X66559
started creating 4 allele had the highest and lowest number of alleles. The mean number of alleles detected
per locus 4.963, respectively. The highest allele frequency of allele 4 position X17031, with a frequency of
0.460. The lowest allele frequency was observed in 0.005 to 6 allele status is X17162. Polymorphism
information content of the lowest 0.839 and 0.703 the primers X80044 and Z66559, respectively, were
observed. Mean PIC obtained for all positions equal to 0.774 in the high heterozygosity of the masses.
Grouped using Nei's genetic similarity algorithm and UPGMA, the masses of the four groups.
