چکیده
ﺗﻨﺶ ﺧﺸﻜﻲ ﻳﻜﻲ از ﻣﻬﻢﺗﺮﻳﻦ ﺗﻨﺶﻫﺎﻳﻲ اﺳﺖ ﻛﻪ ﺳﺒﺐ ﻛﺎﻫﺶ در ﻋﻤﻠﻜﺮد ﮔﻴﺎﻫﺎن زراﻋﻲ ﻣﻲﺷﻮد. اوﻟﻴﻦ ﻣﺮﺣﻠﻪ در ﺳﻴﻜﻞ زﻧﺪﮔﻲ ﮔﻴﺎه،ﺟﻮاﻧﻪزﻧﻲ اﺳﺖ ﻛﻪ ﺑﻪ ﺷﺪت از ﺗﻨﺶ ﺧﺸﻜﻲ آﺳﻴﺐ ﻣﻲﭘﺬﻳﺮد. ﺗﻨﺶ ﺧﺸﻜﻲ ﺑﺮ ﺑﺴﻴﺎری از ﻓﻌﺎﻟﻴﺖﻫﺎی ﻣﻮﻟﻜﻮﻟﻲ از ﺟﻤﻠﻪ ﺗﻐﻴﻴﺮ در اﻟﮕﻮی ﭘﺮوﺗﺌﻮم ﺑﻪ وﻳﮋه در ﻧﺎﺣﻴﺔ ﻣﺤﻮرﺟﻨﻴﻦ ﺑﺬر ﺗﺎﺛﻴﺮ ﺑﺴﺰاﻳﻲ دارد. ﺑﻪ ﻣﻨﻈﻮر ﻓﻬﻢ اﺛﺮ ﺗﻨﺶ ﺧﺸﻜﻲ ﺑﺮ ﺗﻐﻴﻴﺮات ﭘﺮوﺗﺌﻴﻨﻲ ﻃﻲ ﺟﻮاﻧﻪزﻧﻲ،ﻣﻄﺎﻟﻌﻪای در اﻳﻦ راﺳﺘﺎ اﻧﺠﺎم ﺷﺪ. اﺛﺮ ﺗﻨﺶ ﺧﺸﻜﻲ ﺑﺎر( ﺑﺮ اﻟﮕﻮی ﭘﺮوﺗﺌﻴﻨﻲ ﻣﺤﻮر ﺟﻨﻴﻦ ﮔﻨﺪم )ﺳﺮداری و ﻗﺪس ﺑﻪﺗﺮﺗﻴﺐ ﻣﻘﺎوم و ﺣﺴﺎس( ﺑﻪﺻﻮرت ﻓﺎﻛﺘﻮرﻳﻞ در ﻗﺎﻟﺐ ﻃﺮح ﺑﻠﻮک ﻛﺎﻣﻞ ﺗﺼﺎدﻓﻲ در ﺳﻪ ﺗﻜﺮار در ﻓﺎز دوم ﺟﻮاﻧﻪزﻧﻲ ﺑﺮرﺳﻲ ﺷﺪ. ﻧﺘﺎﻳﺞ ﺣﺎﻛﻲ از اﺛﺮات ﻧﺎﻣﻄﻠﻮب ﺗﻨﺶ ﺧﺸﻜﻲ ﺑﺮ درﺻﺪ ﺟﻮاﻧﻪزﻧﻲ ﮔﺰارش ﺷﺪ. ﺗﺠﺰﻳﺔ ژلﻫﺎی اﻟﻜﺘﺮوﻓﻮرز دوﺑﻌﺪی ﻧﺸﺎن داد ﻛﻪ 1000 ﻟﻜﺔ ﺗﻜﺮارﭘﺬﻳﺮ در اﻟﮕﻮی ﭘﺮوﺗﺌﻴﻨﻲ ﻣﺸﺎﻫﺪه ﺷﺪ ﻛﻪ 25 ﻟﻜﻪ در ﺳﻄﺢ 5 درﺻﺪ ﺗﻔﺎوت ﻣﻌﻨﻲداری را ﻧﺸﺎن دادﻧﺪ. ﺑﻪ ﻧﻈﺮ ﻣﻲرﺳﺪ ﻛﻪ ﭘﺮوﺗﺌﻴﻦﻫﺎی اﻓﺰاﻳﺶ ﺑﻴﺎن ﻳﺎﻓﺘﻪ در ارﻗﺎم ﻣﻘﺎوم در ﮔﺮوه ﭘﺮوﺗﺌﻴﻦﻫﺎی ﻣﺮﺗﺒﻂ ﺑﺎ ﺗﻨﺶ و ﻣﺘﺎﺑﻮﻟﻴﺴﻢ ﻛﺮﺑﻮﻫﻴﺪارتﻫﺎ و در ارﻗﺎم ﺣﺴﺎس ﻣﻲﺗﻮاﻧﺪ ﭘﺮوﺗﺌﻴﻦﻫﺎی اﻓﺰاﻳﺶ ﺑﻴﺎن ﻳﺎﻓﺘﻪ ﻣﺮﺗﺒﻂ ﺑﺎ ﺑﺎزدارﻧﺪهﻫﺎی آﻟﻔﺎ آﻣﻴﻼزی ﺑﺎﺷﻨﺪ. ﻧﺘﺎﻳﺞ ﻧﺸﺎن داد ﻛﻪ ﻓﺮاواﻧﻲ اﻳﻦ ﭘﺮوﺗﺌﻴﻦﻫﺎ ﺗﺤﺖ ﺗﻨﺶ ﺧﺸﻜﻲ ﺑﻪ ﺷﺪت و ﻣﻘﺪار ﺗﻨﺶ و زﻣﺎن آﺑﻨﻮﺷﻲ ﺑﺴﺘﮕﻲ دارد.

Proteome analysis of wheat seed embryo (Triticum aestivum) in two tolerance and sensitive cultivars under drought stress in second germination phase

Abstract
Drought stress is one the most important stresses that diminish yield crops. Germination is first phase in plant cycles that damaged from this stress extremely. Drought stress effects many molecular activity as likes changes in proteome patterns in seed embryo axes especially. A study was conducted in order to understand the effects of drought stress on protein changes during germination. To examine the effects of drought stress (-12 bar) on wheat seed embryo (Sardari and Qods respectively tolerance and sensitive cultivars), factorial experimental was conducted in a block complete design with three replications under second germination phase. Results demonstrated that drought stress had inappropriate effects on germination percentage. According to the gels analysis, about 1000 repeatable spots in protein patterns was detected that 25 spots showed significant differences under control and drought stress. It appears that increases in proteins frequency in tolerance and sensitive cultivars related to proteins groups of stresses and carbohydrate metabolism, alpha-amylase inhibitors respectively .The results showed that the frequency of these proteins depends on severity and mount of stress and imbibition time.
